【摘要】：目的研究左舒必利注射液在中國健康受試者單次及多次給藥的藥代動力學。方法用開放、隨機、平行的試驗設計。30名受試者,男女各半,分為3個劑量組,分別接受單次及多次肌內注射不同劑量左舒必利,采集不同時間的血樣,用高效液相色譜-串聯質譜法(HPLC/MS/MS)測定血漿中左舒必利的濃度。用DAS 3.0軟件計算藥代動力學參數。結果單次肌內注射25,50,75 mg左舒必利的主要藥代動力學參數:cmax分別為(724.70±248.91),(949.60±234.80),(1619.00±366.80)μg·L-1;t1/2分別為(7.65±1.32),(7.58±0.89),(8.01±0.88)h,AUC0-t分別為(2874.17±1093.71),(4481.75±913.09),(7559.33±1428.87)μg·L-1·h。連續(xù)給藥組t1/2為(7.41±0.79)h;AUC0-t為(4658.33±909.51)μg·L-1·h。結論中國健康受試者單次肌內注射給藥左舒必利在25~75 mg內呈線性藥代動力學特征,多次給藥沒有蓄積傾向,男、女間比較,差異無統計學意義。
[Abstract]:Objective to study the pharmacokinetics of left sulpiride injection in Chinese healthy volunteers. Methods A total of 30 subjects, half male and half male, were divided into three dose groups, and received intramuscular injection of different doses of levosulpiride at a single and multiple doses, and blood samples were collected at different times. The concentration of levosulpiride in plasma was determined by high performance liquid chromatography-tandem mass spectrometry (HPLC/MS/MS). The pharmacokinetic parameters were calculated by DAS 3.0 software. Results the main pharmacokinetic parameters of single intramuscular injection of left sulpiride at 75 mg were (724.70 鹵248.91), () 949.60 鹵234.80), (1619.00 鹵366.80) 渭 g / L -1 / 2, respectively (7.65 鹵1.32), (7.58 鹵0.89), (8.01 鹵0.88) h AUC0-t were (2874.17 鹵1093.71), (4481.75 鹵913.09), (7559.33 鹵1428.87) 渭 g hL-1 respectively. T 1 / 2 was (7.41 鹵0.79) h ~ (-1) AUC _ (0-t) and (4658.33 鹵909.51) 渭 g 路L ~ (-1) h in continuous administration group. Conclusion the pharmacokinetic characteristics of single intramuscular injection of levosulpiride in Chinese healthy volunteers were linear within 2575 mg, and there was no accumulation tendency in multiple doses. There was no significant difference between male and female.
