鹽酸哌甲酯速溶膜在大鼠體內(nèi)的藥動(dòng)學(xué)研究

【摘要】:目的:研究鹽酸哌甲酯(methylphenidate hydrochloride,MPH)速溶膜(oral fast dissolving films,OFDF)在大鼠體內(nèi)的藥動(dòng)學(xué)。方法:12只大鼠隨機(jī)分為MPH灌胃組(40 mg·kg-1)、MPH-OFDF舌下給藥組(40 mg·kg~(-1))2組,每組6只。麻醉后分別給藥并在不同時(shí)間點(diǎn)取血。采用高效液相色譜法測(cè)定大鼠血漿中MPH濃度,用DAS 2.1軟件計(jì)算藥動(dòng)學(xué)參數(shù),并用SPSS 20.0統(tǒng)計(jì)軟件進(jìn)行獨(dú)立性t檢驗(yàn)分析。結(jié)果:MPH灌胃組t1/2為(1.81±0.81)h,AUC0~4h為(3.49±0.42)mg·L~(-1)·h,tmax和Cmax分別為(0.58±0.13)h和(2.44±0.36)mg·L~(-1)。MPH-OFDF舌下給藥組t1/2為(1.35±0.53)h,AUC0~4h為(15.36±0.53)mg·L~(-1)·h,tmax和Cmax分別為(0.33±0.13)h和(9.49±0.70)mg·L~(-1)。MPH-OFDF舌下組與MPH灌胃組相比,t1/2縮短,但無(wú)顯著性差異,而tmax顯著縮短(P0.01),AUC0~4h和Cmax顯著增大(P0.01)。結(jié)論:與MPH口服給藥相比,MPH-OFDF舌下給藥吸收快,生物利用度有所提高。
[Abstract]:Aim: to study the pharmacokinetics of methylphenidate hydrochloride (methylphenidate hydrochloride,MPH) instant membrane (oral fast dissolving films,OFDF in rats. Methods: twelve rats were randomly divided into MPH group (40 mg kg-1) and sublingual MPH-OFDF group (40 mg kg~ (-1) with 6 rats in each group. After anesthesia, the drug was given separately and blood was taken at different time points.
