【摘要】：皮膚角質(zhì)層是藥物經(jīng)皮滲透的主要障礙,尤其是對(duì)大分子藥物或親水性藥物分子,而微米尺寸的微針可以以微創(chuàng)的方式刺穿皮膚角質(zhì)層,產(chǎn)生可逆的微孔道,促進(jìn)藥物滲透,進(jìn)而克服傳統(tǒng)藥物經(jīng)皮滲透率低的問題。同時(shí),由于微針插入皮膚的深度有限,不觸及皮下痛覺神經(jīng),可以避免引起疼痛。聚合物微針不僅具有足夠的機(jī)械強(qiáng)度刺穿皮膚角質(zhì)層,而且具有優(yōu)異的生物相容性、載藥量可調(diào)節(jié)范圍大等特點(diǎn),即使斷裂在皮膚內(nèi),也不會(huì)產(chǎn)生安全問題。聚合物微針的眾多優(yōu)勢(shì)使其在經(jīng)皮傳遞藥物方面具有廣闊的前景,是當(dāng)前經(jīng)皮給藥領(lǐng)域的研究熱點(diǎn)。本文將從聚合物微針的種類、制備方法、經(jīng)皮給藥應(yīng)用等方面詳細(xì)綜述聚合物微針的最新研究進(jìn)展,并對(duì)這一領(lǐng)域的未來發(fā)展進(jìn)行展望。
[Abstract]:The skin cuticle is a major barrier to drug transdermal penetration, especially for macromolecular drugs or hydrophilic drug molecules, and micron-sized microneedles can penetrate the cuticle of the skin in a minimally invasive manner, producing reversible micropores that promote drug penetration. Thus, the problem of low transdermal permeability of traditional drugs is overcome. At the same time, due to the limited depth of microneedle insertion into the skin, do not touch the subcutaneous pain nerve, can avoid pain. Polymer microneedles not only have enough mechanical strength to penetrate the cuticle of skin, but also have excellent biocompatibility and wide range of drug loading. Even if it breaks in the skin, it will not cause safety problems. Polymer microneedles have a broad prospect in the field of transdermal drug delivery due to their many advantages, which is a hot spot in the field of transdermal drug delivery. In this paper, the latest research progress of polymer microneedles is reviewed in detail from the aspects of kinds, preparation methods, percutaneous drug delivery and so on, and the future development in this field is prospected.
