雷帕霉素互變異構(gòu)體的分離和鑒定

【摘要】:目的分離、純化HPLC圖譜中一直伴隨著雷帕霉素的異構(gòu)體(Rap-T)和它的轉(zhuǎn)化產(chǎn)物并鑒定它們的化學(xué)結(jié)構(gòu)。方法制備液相色譜儀分離Rap-T及其轉(zhuǎn)化產(chǎn)物;HPLC跟蹤分析Rap-T在25℃、50%乙腈-水溶液中不同時間的轉(zhuǎn)化結(jié)果 ;對Rap-T及其轉(zhuǎn)化產(chǎn)物進(jìn)行理化性質(zhì)和波譜分析。結(jié)果 Rap-T是雷帕霉素化學(xué)結(jié)構(gòu)上的吡喃環(huán)擴(kuò)展為氧雜環(huán)庚烷的化合物,其轉(zhuǎn)化產(chǎn)物的化學(xué)結(jié)構(gòu)與雷帕霉素一致;Rap-T與雷帕霉素在25℃、50%乙腈-水溶液中能相互轉(zhuǎn)化。結(jié)論 Rap-T是雷帕霉素的互變異構(gòu)體,二者可以相互轉(zhuǎn)化。
