乙酸陰道熱敏凝膠劑流變性和體外釋藥行為的研究

【摘要】:目的:乙酸陰道熱敏原位凝膠劑(Acetic Acid vaginal thermosensitive in situ gel,AceAcid TISG)作為一種候選陰道外用避孕藥,具有溫度敏感性的特點,本文旨在考察在不同溫度下的流變性質、凝膠溶蝕以及體外釋放特性。方法:冷法制備Ace-Acid TISG,采用Kinexus Pro高級旋轉流變儀測定相轉變溫度,膠凝溫度以及不同溫度下相變過程中的流變參數;無膜溶出法檢測凝膠的溶蝕行為;電位滴定法測定乙酸的體外釋放度。結果:Ace-Acid TISG相轉變溫度為25℃,膠凝溫度為30℃;低溫下為流體,粘彈性很低,相角很高;溫度升高,粘彈性呈指數增長,相角降低。乙酸體外釋放速率與凝膠溶蝕速率呈現良好的線性關系,兩者遵循零級動力學方程。結論:本凝膠制劑具有良好的熱敏感性和體外釋放性,符合陰道即用型凝膠的要求,有望在臨床中獲得應用。
[Abstract]:Objective: acetic acid vaginal thermosensitive in situ gel (Acetic Acid vaginal thermosensitive in situ gel,AceAcid TISG) as a candidate for external use of contraceptive has the characteristics of temperature sensitivity. The purpose of this study was to investigate the rheological properties of contraceptive at different temperatures. Gel dissolution and release characteristics in vitro. Methods: Ace-Acid TISG, was prepared by cold method. Phase transition temperature, gelation temperature and rheological parameters during phase transformation were measured by Kinexus Pro advanced rotary rheometer, and the dissolution behavior of gel was detected by membrane free dissolution method.
