二甲基甲酰胺對(duì)HL7702肝細(xì)胞毒性及線粒體膜電位的影響
[Abstract]:Objective to observe the effects of dimethylformamide (DMF) on the viability, apoptosis and mitochondrial membrane potential of HL7702 hepatocytes, and to explore the possible mechanism of apoptosis induced by DMF. Methods HL7702 hepatocytes were treated with 50100150200 and 250 mmol/L DMF for 24 h. The survival rate and (LDH) release of lactate dehydrogenase were measured. According to the results of cytotoxicity test, control group and DMF group (200 mmol/L) were set up. The effects of DMF on apoptosis of HL7702 cells and mitochondrial membrane potential were detected by flow cytometry. Results DMF decreased the survival rate of HL7702 cells in a dose-dependent manner. The survival rate of HL7702 cells in the 150200 and 250 mmol/L DMF groups was significantly lower than that in the control group, and the content of LDH released to the extracellular cells increased with the increase of the concentration of DMF. Compared with the control group, the apoptosis rate increased significantly (P0.001), the mitochondrial membrane potential decreased and the difference was statistically significant (P0.001). Conclusion the toxic effect of DMF on HL7702 cells is dose-dependent. The change of mitochondrial membrane potential induced by DMF is related to the apoptosis of HL7702 cells induced by DMF.
