輔助分子伴侶對(duì)HSP90構(gòu)象功能的調(diào)節(jié)及其在腫瘤中的作用

【摘要】:熱休克蛋白90(heat shock protein 90,HSP90)作為一種重要的分子伴侶,參與調(diào)控眾多原癌客戶蛋白的折疊、裝配和成熟,在腫瘤的發(fā)生發(fā)展中發(fā)揮重要作用。三磷酸腺苷(adenosine triphosphate,ATP)的結(jié)合和二磷酸腺苷(adenosine diphosphate,ADP)/ATP的交換是驅(qū)動(dòng)HSP90分子伴侶構(gòu)象循環(huán)的關(guān)鍵要素。其中一些輔助分子伴侶協(xié)助HSP90的構(gòu)象循環(huán)過程,參與腫瘤惡性進(jìn)展。本文對(duì)一些常見的輔助分子伴侶,如Hop、CDC37、p23、AHA1和PP5等的結(jié)構(gòu)、功能及其協(xié)助HSP90參與腫瘤發(fā)生發(fā)展的過程進(jìn)行綜述。
[Abstract]:As an important molecular chaperone, heat shock protein 90 (HSP90) is involved in the regulation of the folding, assembly and maturation of many proto-cancer client proteins, and plays an important role in the development of cancer. The binding of adenosine triphosphate (adenosine triphosphate,ATP) and the exchange of adenosine diphosphate (adenosine diphosphate,ADP) / ATP are the key factors driving the conformational cycle of HSP90 molecular chaperones. Some of the accessory molecular chaperones assist in the conformational cycle of HSP90 and participate in the malignant progression of the tumor. In this paper, the structures, functions of some common comolecular chaperones, such as Hop,CDC37,p23,AHA1 and PP5, and the process of assisting HSP90 in tumorigenesis and development are reviewed.
