載G蛋白偶聯(lián)受體APJ的磷脂納米盤的研制

【摘要】:以G蛋白偶聯(lián)受體APJ(即血管緊張素受體AT1相關(guān)的受體蛋白)、膜骨架蛋白(MSP)和磷脂為原料制備載APJ磷脂納米盤。考察了不同投料比(APJ∶MSP分別為1∶2、1∶4和1∶8)和加入Bio-BEADs樹脂(聚苯乙烯二乙烯基苯樹脂)后4℃孵育時(shí)間(2、4和6 h)對(duì)APJ組裝入磷脂納米盤的效率和單體比例的影響。結(jié)果表明,APJ∶MSP采用1∶4,加入Bio-BEADs后4℃孵育2 h,所得的APJ磷脂納米盤直徑約10 nm,高度約5 nm。組裝入該磷脂納米盤的APJ仍具有與其拮抗劑ML221結(jié)合的能力,提示磷脂納米盤能保留APJ的生物活性。
