【摘要】：目的:采用可調激光-微順序注射-閥上實驗室熒光反猝滅法測定硫酸氫氯吡格雷含量。方法:四苯硼鈉與羅丹明B反應,熒光信號猝滅,加硫酸氫氯吡格雷后,其與四苯硼鈉-羅丹明B體系中的四苯硼鈉反應形成更穩(wěn)定的疏水性離子締合物,使羅丹明B重新釋放出熒光,檢測熒光信號的增強程度(ΔF),通過ΔF與硫酸氫氯吡格雷的濃度(C)的回歸方程,計算硫酸氫氯吡格雷含量。結果:熒光信號的ΔF與硫酸氫氯吡格雷C線性回歸方程為ΔF=4 471.7C+12 552,r=0.998 4,線性范圍為0.7~28.2μmol·L~(-1),檢出下限為0.067μmol·L~(-1),重復性RSD為2.9%,加樣回收率在93.0%~99.5%之間。用該方法對市售硫酸氫氯吡格雷片進行測定,結果為97%~100%。結論:該方法可用于測定藥物中硫酸氫氯吡格雷含量。
[Abstract]:Aim: to determine the content of hydroclopidogrel sulfate by fluorescence reverse quenching method. Methods: sodium tetraphenylboron reacted with Rhodamine B, the fluorescence signal was quenched, and after the addition of clopidogrel sulfate, it reacted with sodium tetraphenylboron and sodium rhodamine B to form a more stable hydrophobic ionic association complex. Rhodamine B was rereleased and the enhancement of fluorescence signal was detected (螖 F), was calculated by the regression equation between 螖 F and concentration (C) of hydrogen clopidogrel sulfate), and the content of hydrogen clopidogrel sulfate was calculated. Results: the linear regression equation between 螖 F of fluorescence signal and clopidogrel C sulfate was 螖 F 4471.7C 12552 ru 0.998 4, the linear range was 0.7 渭 mol L ~ (-1), the detection limit was 0.067 渭 mol L ~ (-1), the repeatability RSD was 2.9%, and the recovery rate was between 93.0% and 99.5%. The method was used for the determination of clopidogrel sulfate tablets. Conclusion: this method can be used for the determination of hydroclopidogrel sulfate in drugs.
