十二烷基硫酸鈉純度對(duì)坎地沙坦酯溶出的影響

【摘要】:目的:研究十二烷基硫酸鈉(SDS)純度對(duì)坎地沙坦酯溶出的影響,揭示SDS自身純度及含有混合物的比例對(duì)藥物溶出度檢驗(yàn)結(jié)果的影響。方法:在SDS中加入0.5 mol·L-1硫酸溶液25m L回流后用乙醚提取,將SDS轉(zhuǎn)化為十二烷醇,采用GC法進(jìn)行測(cè)定。GC法:色譜柱HP-5(30 mm×0.53 mm,2.65μm),進(jìn)樣口溫度270℃,FID檢測(cè)器溫度300℃,載氣為高純氮,流速3.0 m L·min-1,分流比10∶1,升溫程序(起始溫度80℃,保持1 min后,以10℃·min-1的升溫速率升至260℃,保持3 min),進(jìn)樣1μL。結(jié)果:選取具有代表性的4批SDS配制溶液,對(duì)坎地沙坦酯的過(guò)飽和溶液進(jìn)行了測(cè)定,質(zhì)量濃度分別為6.28×10-3、11.59×10-3、6.17×10-3和23.36×10-3mg·m L-1。結(jié)論:SDS的助溶能力與自身純度及含有混合物碳鏈的長(zhǎng)短有關(guān);不同企業(yè)生產(chǎn)的SDS自身純度相同時(shí),含有混合物的碳鏈越長(zhǎng),助溶能力增加,溶出效果相對(duì)更好。
