【摘要】：由于具有靶向性高、不良反應少等優(yōu)勢,治療性抗體已成為生物技術產(chǎn)業(yè)的重要研究領域。諸多重大疾病如腫瘤、自身免疫性疾病、心血管疾病、糖尿病、老年性癡呆等都有望用抗體藥物進行治療,以抗體為基礎的分子設計和靶向治療也是近年生物醫(yī)藥產(chǎn)業(yè)的最大增長點。但目前在抗體藥物的研發(fā)和生產(chǎn)過程中仍然存在一些問題亟待解決。其中,抗體分子聚集是影響藥物成藥性的關鍵因素之一,篩選和設計穩(wěn)定高效的抗體藥物是藥物研發(fā)早期階段的重要目標。相較于傳統(tǒng)篩選方法,應用計算機模擬技術預測蛋白質(zhì)的聚集傾向為抗體藥物的穩(wěn)定性和高度可溶性提供了重要參考信息,將有效地提高藥物的研發(fā)效率。本文重點介紹了已成功應用于治療性抗體領域的聚集預測方法,并對這些方法的原理與應用前景進行總結(jié),為基于計算機模擬的抗體研究提供參考。
[Abstract]:Because of the advantages of high targeting and less adverse reactions, therapeutic antibodies have become an important research field in biotechnology industry. Many major diseases, such as tumors, autoimmune diseases, cardiovascular diseases, diabetes, Alzheimer's disease, and so on, are expected to be treated with antibody drugs. Antibody-based molecular design and targeted therapy are also the biggest growth point of biomedical industry in recent years. However, there are still some problems to be solved in the development and production of antibody drugs. Among them, the aggregation of antibody molecules is one of the key factors affecting the drug formation. Screening and designing stable and efficient antibody drugs is an important goal in the early stage of drug development. Compared with the traditional screening methods, the application of computer simulation technology to predict the aggregation tendency of proteins provides important reference information for the stability and high solubility of antibody drugs, which will effectively improve the efficiency of drug research and development. This paper mainly introduces the clustering prediction methods which have been successfully applied in the field of therapeutic antibodies, and summarizes the principles and application prospects of these methods, which provides a reference for the study of antibodies based on computer simulation.
