【摘要】：目的:探討左卡尼汀對(duì)糖尿病心肌病模型大鼠心肌細(xì)胞凋亡的影響。方法:采用高脂高糖飼料加腹腔注射鏈脲佐菌素建立糖尿病心肌病大鼠模型,隨機(jī)分為模型(生理鹽水)組和左卡尼汀低、中、高劑量(30、40、50 mg/d)組,另取正常大鼠作為正常對(duì)照(純凈水)組,每組10只,ig給予相應(yīng)藥物,每天1次,連續(xù)6周。末次給藥后檢測(cè)各組大鼠心室內(nèi)壓、心肌細(xì)胞凋亡指數(shù)和心肌細(xì)胞中B淋巴細(xì)胞瘤2(Bcl-2)、Bcl-2相關(guān)X蛋白(Bax)、p53基因蛋白的表達(dá)。結(jié)果:與正常對(duì)照組和模型組比較,左卡尼汀低、中、高劑量組大鼠的心率、左心室收縮壓、左心室內(nèi)壓最大上升和下降速率均降低,左心室舒張末期壓升高,差異具有統(tǒng)計(jì)學(xué)意義(P0.01);與模型組比較,左卡尼汀低、中、高劑量組大鼠的心肌細(xì)胞凋亡指數(shù)、Bax和p53基因蛋白的表達(dá)均明顯降低,Bcl-2基因蛋白的表達(dá)明顯升高,差異具有統(tǒng)計(jì)學(xué)意義(P0.01或P0.05),其中以左卡尼汀中劑量組效果最顯著。結(jié)論:左卡尼汀對(duì)糖尿病心肌病模型大鼠心肌細(xì)胞的凋亡有抑制作用。
[Abstract]:Aim: to investigate the effect of levocarnitine on myocardial apoptosis in diabetic cardiomyopathy rats. Methods: the diabetic cardiomyopathy rats were established by high fat and high glucose diet plus intraperitoneal injection of streptozotocin. The rats were randomly divided into three groups: the model group (normal saline) and the low, medium and high dose group (30 carnitine 50 mg/d). Normal rats were taken as normal control (pure water) group. 10 rats in each group were given corresponding drugs once a day for 6 weeks. After the last administration, intraventricular pressure, myocardial apoptosis index and the expression of B lymphocyte tumor 2 (Bcl-2) Bcl-2 related X protein (Bax) p53 gene were detected in each group. Results: compared with the normal control group and the model group, the heart rate, left ventricular systolic pressure, maximum rise and decrease rate of left ventricular pressure and left ventricular end-diastolic pressure increased in the middle and high dose groups. The difference was statistically significant (P0.01), compared with the model group, the expression of Bcl-2 gene protein and Bax gene protein were significantly decreased in the high dose and middle dose groups. The difference was statistically significant (P0.01 or P0.05), in which L-carnitine medium dose group had the most significant effect. Conclusion: L-carnitine can inhibit cardiomyocyte apoptosis in diabetic cardiomyopathy rats.
