【摘要】：目的:建立人血清中茶堿、二羥丙茶堿和多索茶堿血藥濃度同時測定的高效液相色譜法并應(yīng)用于患者的個體化治療。方法:采用蛋白沉淀法對血樣進(jìn)行預(yù)處理,以咖啡因?yàn)閮?nèi)標(biāo)。色譜條件:Eclipse XDB-C18(4.6 mm×150 mm,5μm)色譜柱,流動相為甲醇-水梯度洗脫,流速為1.0 ml·min-1,檢測波長273 nm,柱溫為30℃。結(jié)果:茶堿在1.47~47.00μg·ml-1范圍內(nèi)線性良好(r=0.998),二羥丙茶堿在1.28~40.80μg·ml-1范圍內(nèi)線性良好(r=0.999),多索茶堿在1.44~46.00μg·ml-1范圍內(nèi)線性良好(r=0.999)。茶堿、二羥丙茶堿和多索茶堿的平均絕對回收率均達(dá)到95%左右,平均相對回收率分別為100.34%~105.48%,97.09%~103.13%和97.61%~101.39%,日內(nèi)和日間RSD均小于10%。結(jié)論:本方法簡便、快速、靈敏,可用于茶堿、二羥丙茶堿和多索茶堿的臨床血藥濃度監(jiān)測及藥動學(xué)研究。
[Abstract]:Objective: to establish a high performance liquid chromatography (HPLC) for simultaneous determination of theophylline, dihydroxypropylline and doxofylline in human serum.
