中空性納米聚集顆粒用于肺部給藥的研究進(jìn)展

【摘要】:肺部給藥是一種簡(jiǎn)單、直接、有效、無(wú)侵害性的給藥方式,肺部藥物遞送可以達(dá)到局部及全身的治療效果。納米給藥系統(tǒng)具有靶向及緩釋特性,在提高治療指數(shù)、降低不良反應(yīng)方面具有顯著優(yōu)勢(shì)。通過(guò)將納米粒溶液或納米粒和輔料的混合液進(jìn)行噴霧干燥,可制得中空性納米聚集顆粒。中空性納米聚集顆粒具有較大幾何直徑和較小的密度,能夠提高粒子的空氣動(dòng)力學(xué)直徑,得到良好的肺部沉積率,是一種潛在的肺部納米給藥系統(tǒng)新劑型。本文從中空性納米聚集顆粒的形成原理、影響因素、制備工藝等方面闡述了該新型制劑的關(guān)鍵技術(shù),并對(duì)其物理化學(xué)性質(zhì)、粉體動(dòng)力學(xué)性質(zhì)以及用于肺部給藥的評(píng)價(jià)指標(biāo)進(jìn)行了總結(jié),為該種新劑型應(yīng)用于肺部給藥的干粉吸入劑提供了制劑學(xué)參考。
[Abstract]:Pulmonary drug delivery is a simple, direct, effective and non-invasive drug delivery method. Pulmonary drug delivery can achieve local and systemic therapeutic effects. Nano-drug delivery system has the characteristics of targeting and slow release, and has significant advantages in improving the treatment index and reducing adverse reactions. Hollow nanoparticles can be prepared by spray drying of nanoparticles or mixtures of nanoparticles and excipients. Hollow nanoparticles have a large geometric diameter and a small density, which can improve the aerodynamics diameter of particles and obtain a good lung deposition rate. It is a potential new dosage form of pulmonary nano-drug delivery system. In this paper, the key technology of the new preparation is described from the forming principle, influencing factors and preparation process of the hollow nano-aggregate particles, and the physical and chemical properties of the new preparation are also discussed. The kinetic properties of the powder and the evaluation indexes used in pulmonary administration were summarized, which provided the preparation reference for the application of the new dosage form in lung administration of dry powder inhalants.
