液相色譜-串聯(lián)質(zhì)譜法測(cè)定大鼠血漿中舒巴坦的濃度及其藥動(dòng)學(xué)研究

【摘要】:目的:建立測(cè)定大鼠血漿中舒巴坦血藥濃度的液相色譜-串聯(lián)質(zhì)譜(LC-MS/MS)法,探討美羅培南、亞胺培南對(duì)舒巴坦在大鼠體內(nèi)的藥動(dòng)學(xué)影響。方法:將18只SD大鼠隨機(jī)分為舒巴坦組、美羅培南+舒巴坦組和亞胺培南+舒巴坦組,分別靜脈注射給藥,按規(guī)定時(shí)間點(diǎn)采血,血漿樣品經(jīng)乙酸乙酯萃取,LC-MS/MS法測(cè)定血藥濃度;采用負(fù)離子模式,多重反應(yīng)監(jiān)測(cè)(multiple reaction monitoring,MRM),定量離子對(duì)為m/z232.3→m/z139.9(舒巴坦)和m/z423.4→m/z 207.1(頭孢呋辛,內(nèi)標(biāo))。經(jīng)DAS 2.0.1計(jì)算藥動(dòng)學(xué)參數(shù),并進(jìn)行統(tǒng)計(jì)學(xué)比較。結(jié)果:舒巴坦在0.250~200μg·mL~(-1)范圍內(nèi)線性關(guān)系良好,方法學(xué)符合要求。藥動(dòng)學(xué)試驗(yàn)結(jié)果表明,合用美羅培南或亞胺培南后,舒巴坦在大鼠體內(nèi)的主要藥動(dòng)學(xué)參數(shù)t_(max)、C_(max)、AUC、MRT、t_(1/2z)、CL_z和V_z與單用舒巴坦相比,無顯著性差異。結(jié)論:和已報(bào)道的方法比較,本研究所建方法具有快速、高效,生物樣品用量小的特點(diǎn),適用于舒巴坦在大鼠體內(nèi)藥動(dòng)學(xué)的研究;美羅培南、亞胺培南對(duì)舒巴坦在大鼠體內(nèi)的藥動(dòng)學(xué)過程無明顯影響,提示評(píng)價(jià)此類藥物相互作用還需同時(shí)考慮藥效學(xué)等方法。
[Abstract]:Aim: to establish a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the determination of sulbactam in rat plasma and to investigate the pharmacokinetic effects of meropenem and imipenem on sulbactam in rats. Methods: eighteen SD rats were randomly divided into three groups: sulbactam group, meropenem sulbactam group and imipenem sulbactam group. The quantitative ion pairs were m/z232.3 m/z139.9 and m/z423.4 m / z 207.1 (cefuroxime, internal standard). Pharmacokinetic parameters were calculated by DAS 2. 0. 1 and compared statistically. Results: the linear range of sulbactam was 0.250 渭 g mL ~ (-1). The results of pharmacokinetic test showed that there was no significant difference between sulbactam and sulbactam compared with sulbactam alone in the pharmacokinetics of sulbactam in rats. Conclusion: compared with the reported method, this method has the characteristics of fast, high efficiency and small amount of biological sample, which is suitable for the pharmacokinetics of sulbactam in rats. Imipenem had no obvious effect on the pharmacokinetic process of sulbactam in rats.
