藥根堿與DNA G-四鏈體的相互作用研究

【摘要】:DNA單鏈(PS2.M)在K~+存在下可被誘導(dǎo)形成G-四鏈體。文章利用圓二色譜(CD)以及核磁共振光譜(NMR)表征了G-四鏈體的形成,并通過熒光滴定光譜、紫外滴定光譜以及核磁共振光譜研究了鹽酸藥根堿與G-四鏈體的相互作用。加入G-四鏈體后,藥根堿在530nm處的熒光明顯增強(qiáng);紫外吸收光譜表現(xiàn)出減色和紅移現(xiàn)象,說明藥根堿可能堆積到G-四鏈體的尾部或者插入到G-四鏈體尾部的兩個(gè)G-四分體之間形成夾心結(jié)構(gòu)。由等摩爾連續(xù)變化法算出藥根堿和DNA的結(jié)合比為1∶1,利用Scatchard方程計(jì)算出二者的結(jié)合常數(shù)為3.12×10~6 L/mol、結(jié)合位點(diǎn)為0.98。實(shí)驗(yàn)表明:鹽酸藥根堿與G-四鏈體有很強(qiáng)的相互作用,藥根堿可以穩(wěn)定DNA的G-四鏈體結(jié)構(gòu),二者的作用模式可能為末端堆積。
[Abstract]:DNA single strand (PS2.M) can be induced to form G-4 chain in the presence of K ~. The formation of G- quaternary chain was characterized by circular dichroism (CD) and nuclear magnetic resonance spectroscopy (NMR). The interaction between drug base hydrochloride and G- quaternary was studied by fluorescence titration, ultraviolet titration and nuclear magnetic resonance spectroscopy. After the addition of G-quadruplex, the fluorescence of the root alkaloids at 530nm was obviously enhanced, and the UV absorption spectrum showed the phenomenon of decreasing color and red shift. It is suggested that the nucleotide alkaloids may be stacked at the tail of the G- quadruplex or intercalated into the tail of the G- quaternary to form a sandwich structure between the two G- tetrads. The binding ratio of root alkaloid to DNA is 1: 1, the binding constant is 3.12 脳 10 ~ (6) L / mol and the binding site is 0.98 by Scatchard equation. The results show that there is a strong interaction between the drug root base and the G-quaternary chain, and the drug root alkaloids can stabilize the structure of the G-quaternary chain of DNA. The interaction pattern between them may be terminal stacking.
