【摘要】：目的制備一種工藝簡便、易于放大生產(chǎn)的吲哚布芬緩釋微丸。方法結(jié)合流化床的底噴技術(shù)與液相層積法進(jìn)行空白丸芯上藥制備載藥微丸,以新型水分散體Kollicoat~汶SR30D為包衣材料制備緩釋微丸,考察包衣處方因素、熱處理?xiàng)l件和釋放介質(zhì)等因素對釋放度的影響。采用BET比表面積分析儀測定衣膜孔隙度和比表面積,探討致孔劑對衣膜孔隙度的影響機(jī)制。初步考察微丸在不同條件下的穩(wěn)定性并推測其體外的釋放機(jī)制。結(jié)果吲哚布芬緩釋微丸圓整度良好,包衣膜光滑完整,載藥質(zhì)量分?jǐn)?shù)約為50%,在加速試驗(yàn)和長期試驗(yàn)中均能保持穩(wěn)定的釋藥特性。致孔劑的添加能夠增加衣膜內(nèi)部的孔容及孔隙數(shù)量,從而顯著增加膜層的比表面積。微丸體外釋藥機(jī)制符合一級動力學(xué)。結(jié)論所制得的吲哚布芬緩釋微丸釋藥平穩(wěn)、性質(zhì)穩(wěn)定、制備工藝簡便、易于操作。
[Abstract]:Objective to prepare an indole-ibufen sustained-release pellet which is simple and easy to be amplified. Methods combined with the bottom spray technique of fluidized bed and the liquid phase stratification method, the drug loaded pellets were prepared by using the blank pellet core and the new water dispersion Kollicoatine SR30D as the coating material. The factors of coating prescription were investigated. The effect of heat treatment conditions and release medium on the release degree. The porosity and specific surface area of the coating film were measured by BET surface area analyzer, and the mechanism of the effect of pore-forming agent on the porosity of the coating film was discussed. The stability of pellets under different conditions was investigated and the release mechanism of pellets in vitro was inferred. Results the indole ibufen sustained release pellets had a good roundness, a smooth coating film, a drug loading mass fraction of about 50%, and stable release characteristics in both accelerated and long-term tests. The addition of pore-forming agent can increase the pore volume and the number of pores in the film, thus increasing the specific surface area of the film. The drug release mechanism of pellets in vitro accords with the first order kinetics. Conclusion the indole ibufen sustained-release pellets are stable, simple and easy to operate.
