【摘要】:篩選和開(kāi)發(fā)新型低毒的鈣調(diào)磷酸酶抑制劑(CNI)宜揚(yáng)長(zhǎng)避短,充分利用各方法的優(yōu)勢(shì)。本文綜述了酵母"正篩選"模型,酵母報(bào)告基因高通量CNI篩選模型和鈣調(diào)磷酸酶(Ca N)活性檢測(cè)法3種常用的篩選方法的適用范圍、優(yōu)缺點(diǎn)以及多方法聯(lián)合。其中,酵母"正篩選"模型利用生長(zhǎng)圈來(lái)代替抑菌圈,有效避免了抑菌化合物造成的"假陽(yáng)性";酵母報(bào)告基因的高通量篩選模型有較高的篩選效率,適合于大量樣品的初步篩選;Ca N活性檢測(cè)法具有作用靶點(diǎn)明確和抑制效率直觀準(zhǔn)確的優(yōu)點(diǎn)。本文旨在為篩選和開(kāi)發(fā)新型、低毒的CNI提供參考。
[Abstract]:Screening and developing a new type of low toxicity calmodulase inhibitor (CNI) should make full use of the advantages of each method. In this paper, the application, advantages and disadvantages of three common screening methods, including yeast "positive screening" model, yeast reporter gene high-throughput CNI screening model and calcium phosphatase (Ca _ N) activity test, were reviewed. Among them, yeast "positive screening" model uses growth circle instead of bacteriostasis circle, which effectively avoids "false positive" caused by bacteriostatic compounds, and the high-throughput screening model of yeast reporter gene has higher screening efficiency. The primary screening method for the detection of Ca N activity is suitable for a large number of samples. It has the advantages of clear targets and direct and accurate inhibition efficiency. The purpose of this paper is to provide a reference for screening and developing new, low-toxicity CNI.
