【摘要】：目的:建立快速測(cè)定人血漿中酒石酸美托洛爾濃度的方法。方法:血漿樣品以3-叔丁基甲醚處理后,采用高效液相色譜法進(jìn)樣測(cè)定,色譜柱為Kromasil LC-18DB,流動(dòng)相為乙腈-10 mmol/L辛烷磺酸鈉溶液(p H=2.0)(30∶70),柱溫為25℃,流速為1.3ml/min,熒光檢測(cè)波長為267 nm(激發(fā)波長)、290 nm(發(fā)射波長),內(nèi)標(biāo)為吲哚洛爾。結(jié)果:酒石酸美托洛爾血藥濃度在2~300 ng/ml范圍內(nèi)線性關(guān)系良好(r=0.999 9),最低檢測(cè)濃度為1 ng/ml。平均方法回收率為(101.13±4.0)%,日內(nèi)RSD≤2.47%,日間RSD≤4.51%。結(jié)論:該方法簡便、快速、靈敏、重現(xiàn)性好,適用于酒石酸美托洛爾臨床血藥濃度監(jiān)測(cè)及人體藥動(dòng)學(xué)研究。
