異常黑膽質(zhì)成熟劑延緩衰老作用機(jī)制的研究

【摘要】:目的:分別觀察異常黑膽質(zhì)成熟劑(ASMq)對(duì)D-半乳糖所致衰老模型大鼠學(xué)習(xí)記憶能力、血液生化指標(biāo)、免疫功能,心、腦、肝臟組織形態(tài)學(xué)和心、腦、肝組織中p16蛋白、p-半乳糖苷酶表達(dá)以及脾臟NF-κBp65表達(dá)的影響,以此進(jìn)一步闡明ASMq延緩衰老的部分作用機(jī)制。方法:選取3個(gè)月齡雄性SD大鼠60只,體質(zhì)量180~220g,采用D-半乳糖制作亞急性衰老模型,在模型建立后分別用高、中、低劑量(6.0,3.0,1.5g/kg) ASMq對(duì)衰老大鼠模型進(jìn)行干預(yù),并于第67d處死大鼠,測(cè)定各組主要血常規(guī)、血液生化指標(biāo),測(cè)定各組大鼠血清IL-2、1L-6、TNF-α水平,觀察各組大鼠心臟、肝臟、腦組織形態(tài)學(xué)變化和p16蛋白和β-半乳糖苷酶表達(dá)水平以及用半定量RT-PCR法檢各組大鼠脾臟NF-κBp65mRNA表達(dá)水平。結(jié)果:(1)與衰老模型組大鼠相比,正常對(duì)照組抵達(dá)終點(diǎn)的游泳時(shí)間差異有統(tǒng)計(jì)學(xué)意義(p0.01);衰老模型組與ASMq各給藥組組大鼠抵達(dá)終點(diǎn)的游泳時(shí)間差異有統(tǒng)計(jì)學(xué)意義(P0.05-0.01)。(2)衰老模型組與ASMq高、中、低劑量組白細(xì)胞(WBC)、紅細(xì)胞(RBC)、血紅蛋白(HGB)、紅細(xì)胞壓積(HCT)、平均紅細(xì)胞體積(MCV)、血小板數(shù)目(PLT)、谷丙轉(zhuǎn)氨酶(ALT)、谷草轉(zhuǎn)氨酶(AST)、總蛋白(TP)、血肌酐(CRE)、血尿酸(UA)、白蛋白(ALB)、血糖(GLU)差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05),但甘油三酯(TG)、總膽固醇(TC)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)差異有統(tǒng)計(jì)學(xué)意義(P0.05)。(3)與正常對(duì)照組比較,衰老模型組IL-2、IL-6、TNF-α含量差異有統(tǒng)計(jì)學(xué)意義(P0.01);與衰老模型組比較,ASMq高劑量組IL-2含量升高,IL-6、TNF-a含量降低,差異有統(tǒng)計(jì)學(xué)意義(P0.01);ASMq中劑量組IL-2含量升高,IL-6、TNF-α含量降低,差異有統(tǒng)計(jì)學(xué)意義(P0.05-0.01);ASMq低劑量組IL-2含量升高,IL-6含量降低,差異有統(tǒng)計(jì)學(xué)意義(P0.05),TNF-α含量降低,但差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。(4)心臟:與衰老模型組相比,ASMq各給藥組組心肌細(xì)胞飽滿,肌絲排列整齊,但也出現(xiàn)了輕度血管擴(kuò)張;腦:與衰老模型組相比,ASMq高、中劑量組細(xì)胞形態(tài)結(jié)構(gòu)基本完整,排列整齊,但也有輕度的細(xì)胞腫脹,ASMq高、中劑量組細(xì)胞形態(tài)結(jié)構(gòu)基本完整,排列整齊,但也出現(xiàn)了不同程度的的細(xì)胞腫脹。ASMq低劑量組出現(xiàn)了少量的空泡樣變化和核固縮;肝臟:與衰老模型組相比,ASMq各給藥組肝細(xì)胞結(jié)構(gòu)完整,排列整齊。有少量的脂質(zhì)空泡和水腫,但沒(méi)有出現(xiàn)核固縮。中、低劑量出現(xiàn)了脂質(zhì)空泡和水腫,低劑量出現(xiàn)了核固縮(5)各組大鼠心臟、肝臟、腦組織中p16蛋白、β-半乳糖苷酶表達(dá)陽(yáng)性的細(xì)胞表現(xiàn)為細(xì)胞漿染成黃褐色。與正常對(duì)照組比較,衰老模型組心、腦、肝組織中的p16蛋白、β-半乳糖苷酶的表達(dá)明顯升高,差異有統(tǒng)計(jì)學(xué)意義(P0.05~0.01);與衰老模型組比較,ASMq高、中、低劑量組心、腦、肝組織中p16蛋白、β-半乳糖苷酶的表達(dá)明顯下降,差異有統(tǒng)計(jì)學(xué)意義(P0.05~0.01)。(6)與正常對(duì)照組比較,衰老模型組大鼠脾臟NF-KBp65mRNA表達(dá)水平明顯升高,差異有統(tǒng)計(jì)學(xué)意義(P0.01);與衰老模型組比較,ASMq高、中劑量組脾臟NF-KBp65mRNA表達(dá)水平降低,差異有統(tǒng)計(jì)學(xué)意義(P0.05-0.01),低劑量組脾臟NF-KBp65mRNA表達(dá)水平降低,但差異沒(méi)有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:本實(shí)驗(yàn)得出異常黑膽質(zhì)成熟劑具有延緩機(jī)體衰老作用,其作用機(jī)理可能是降低p16蛋白、β-半乳糖苷酶的表達(dá)以及下調(diào)NF-KBp65mRNA表達(dá)水平有關(guān)。
[Abstract]:Objective : To investigate the effects of ASMq on the learning and memory ability , blood biochemical index , immune function , heart , brain , liver tissue morphology and the expression of NF - 魏B P65 in rats with D - galactose induced by D - galactose .
There was no significant difference in swimming time between aging model group and ASMq group ( P0.05 - 0.01 ) . ( 2 ) There was no significant difference between aging model group and ASMq group ( P0.05 ) , but triglyceride ( TG ) , total cholesterol ( TC ) , high density lipoprotein ( HDL ) and low density lipoprotein ( LDL ) were statistically significant ( P0.05 ) .
Compared with the aging model group , the levels of IL - 2 , IL - 6 and TNF - a in ASMq high - dose group were decreased , and the difference was significant ( P0.01 ) .
The levels of IL - 2 , IL - 6 and TNF - 偽 in ASMq decreased significantly ( P0.05 - 0.01 ) .
The levels of IL - 2 and IL - 6 in the low - dose ASMq group were higher than those in the ASMq low - dose group ( P0.05 ) , but there was no statistical significance ( P0.05 ) .
Brain : Compared with the aging model group , ASMq was high , the morphological structure of medium - dose group was basically intact , the arrangement was orderly , but also mild cell swelling , ASMq was high , the morphological structure of medium - dose group was basically intact , the arrangement was orderly , but also the cell swelling of different degree appeared .
