【摘要】：目的:標(biāo)定并評價(jià)人垂體促黃體生成激素(LH)國際候選標(biāo)準(zhǔn)品(批號81/535)的穩(wěn)定性,為WHO評估該候選品作為第3次國際標(biāo)準(zhǔn)品的適用性提供數(shù)據(jù)支持。方法:以第2次LH國際標(biāo)準(zhǔn)品(批號80/552)為標(biāo)準(zhǔn)品,采用放射免疫法、酶聯(lián)免疫法、化學(xué)發(fā)光免疫法和時(shí)間分辨免疫熒光法分析LH國際候選標(biāo)準(zhǔn)品(批號81/535)的效價(jià)和穩(wěn)定性。結(jié)果:本實(shí)驗(yàn)室上報(bào)數(shù)據(jù):LH(批號81/535)效價(jià)的幾何均數(shù)為34.2 IU·安瓿~(-1),相當(dāng)于WHO報(bào)告中提供數(shù)據(jù)的103.0%;穩(wěn)定性樣本B(37℃放置26年)和樣本F(4℃放置26年)效價(jià)的幾何均數(shù)分別為24.8 IU·安瓿~(-1)和32.4 IU·安瓿~(-1)。結(jié)論:全球11個(gè)實(shí)驗(yàn)室提交了數(shù)據(jù)。經(jīng)標(biāo)定,LH(81/535)效價(jià)為33.2IU·安瓿~(-1)(95%可信限為32.1~34.3 IU·安瓿~(-1)),且穩(wěn)定性滿足要求。經(jīng)WHO生物標(biāo)準(zhǔn)專家委員會審核通過,最終確定81/535可作為第3次LH國際標(biāo)準(zhǔn)品使用,每安瓿效價(jià)定為33IU。
[Abstract]:Aim: to calibrate and evaluate the stability of human pituitary luteinizing hormone (LH) candidate standard (batch number 81 / 535) and provide data support for WHO to evaluate the suitability of the candidate as the third international standard.
