【摘要】：目的制備一種以凝溶膠蛋白(GSN)為靶點(diǎn)的包裹液態(tài)氟碳的納米級(jí)PLGA高分子造影劑(GSN-PLGA-PFP),并評(píng)價(jià)其體外熱致相變、靶向及顯影能力。方法采用雙乳化法及碳二亞胺法制備靶向相變型高分子超聲造影劑。檢測(cè)此造影劑的一般性質(zhì);顯微鏡加熱板法進(jìn)行熱致相變;激光共聚焦顯微鏡體外觀察GSN-PLGA-PFP被小鼠腹水型肝癌高淋巴道轉(zhuǎn)移Hca-F細(xì)胞攝取的情況,初步評(píng)價(jià)其尋靶能力;體外低強(qiáng)度聚焦超聲(LIFU)儀輻照后觀察其超聲顯影效果。結(jié)果 GSN-PLGA-PFP的平均粒徑為(328.59±3.82)nm,造影劑表面平均Zeta電位為(-10.36±1.34)mV,造影劑表面GSN單抗連接率高達(dá)98.31%,且于43.5℃時(shí)納米粒開(kāi)始發(fā)生相變;體外攝取實(shí)驗(yàn)顯示Hca-F細(xì)胞可較多地?cái)z取GSNPLGA-PFP,體外行LIFU輻照后可觀察到顯影效果明顯增強(qiáng)。結(jié)論自制攜Gelsolin單抗包載PFP的PLGA造影劑熱致相變效果明顯,可與Hca-F細(xì)胞特異度結(jié)合且體外顯影效果較好。
[Abstract]:Aim to prepare a nano-PLGA high molecular contrast agent (GSN-PLGA-PFP) coated with liquid fluorocarbon (GSN) and evaluate its thermal phase transition, targeting and developing ability in vitro. Methods two-emulsification and carbodiimide methods were used to prepare phase-specific polymer ultrasound contrast agent. The general properties of the contrast medium, the thermo-induced phase change by microscope heating plate, the uptake of GSN-PLGA-PFP by mouse ascites hepatoma hyperlymphoid metastasis Hca-F cells were observed by laser confocal microscopy in vitro, and the targeting ability of GSN-PLGA-PFP by Hca-F cells was preliminarily evaluated. The effect of low intensity focused ultrasound (LIFU) was observed after irradiation. Results the average particle size of GSN-PLGA-PFP was (328.59 鹵3.82) nm, and the average Zeta potential was (-10.36 鹵1.34) MV. In vitro uptake assay showed that Hca-F cells could absorb more GSNPLGA-PFP.After LIFU irradiation in vitro, the effect of GSNPLGA-PFP was obviously enhanced. Conclusion the thermo-induced phase change of PLGA contrast agent with Gelsolin monoclonal antibody encapsulated with PFP is obvious, and it can bind to Hca-F cells specifically and develop well in vitro.
