【摘要】：目的:為進(jìn)一步研究氯胺酮在成癮動(dòng)物體內(nèi)代謝動(dòng)力學(xué)規(guī)律及細(xì)胞色素P_(450)活性的調(diào)控和相關(guān)性研究提供參考。方法:以"細(xì)胞色素P_(450)""性別""氯胺酮""CYP2B""CYP3A""Sex""Ketamine"等為關(guān)鍵詞,組合查詢1980-2016年在Pub Med、Elsevier、中國(guó)知網(wǎng)、萬(wàn)方、維普等數(shù)據(jù)庫(kù)中的相關(guān)文獻(xiàn),對(duì)CYP2B和CYP3A參與藥物代謝的誘導(dǎo)、抑制和性別等影響因素及對(duì)氯胺酮代謝的影響進(jìn)行綜述。結(jié)果與結(jié)論:共檢索到相關(guān)文獻(xiàn)256篇,其中有效文獻(xiàn)62篇。CYP2B和CYP3A是重要的細(xì)胞色素P_(450),二者參與代謝了臨床大多數(shù)藥物,包括氯胺酮。誘導(dǎo)、抑制和性別差異等因素主要是通過(guò)改變細(xì)胞色素P_(450)的基因表達(dá)和蛋白表達(dá)來(lái)影響藥物在動(dòng)物體內(nèi)代謝的。氯胺酮是一種被廣泛濫用的藥物,具有成癮性和個(gè)體的耐受差異性。通過(guò)研究細(xì)胞色素P_(450)的基因表達(dá)和蛋白表達(dá)有利于指導(dǎo)研究氯胺酮在成癮動(dòng)物體內(nèi)的特殊代謝動(dòng)力學(xué)規(guī)律,以及為闡釋成癮動(dòng)物體內(nèi)的細(xì)胞色素P_(450)活性的調(diào)控機(jī)制和相關(guān)性研究提供理論支持。
[Abstract]:Aim: to provide a reference for further study on the regulation and correlation of Ketamine metabolism and cytochrome P450 activity in addictive animals. Methods: using "cytochrome P450", "sex", "ketamine", "CYP2B", "CYP3A", "Sex" Ketamine "as the key words, the relevant literatures in the database of Pub Meden Elsevier, China Zhiwang, Wanfang, Wiper, etc. from 1980 to 2016 were used to investigate the induction of CYP2B and CYP3A involved in drug metabolism. The influence factors such as inhibition and sex and the effect on ketamine metabolism were reviewed. Results & conclusion: a total of 256 literatures were retrieved, of which 62 were effective. CYP2B and CYP3A were important cytochrome P450. They were involved in the metabolism of most clinical drugs, including ketamine. Factors such as induction, inhibition and sex difference affect drug metabolism in animals by changing the gene expression and protein expression of cytochrome P450. Ketamine is a widely abused drug with addictive and individual tolerance. By studying the gene expression and protein expression of cytochrome P450, it is helpful to study the specific metabolic kinetics of ketamine in addictive animals. It also provides theoretical support for the study on the regulation mechanism and correlation of cytochrome P450 activity in addictive animals.
