白藜蘆醇衍生物的合成及其初步細(xì)胞活性研究

【摘要】:目的:合成白藜蘆醇的衍生物--乙酰白藜蘆醇和甲基化白藜蘆醇,并對(duì)其進(jìn)行結(jié)構(gòu)解析;建立它們的液相色譜測(cè)定含量方法并測(cè)定脂水分配系數(shù);觀(guān)察它們對(duì)3T3-L1脂肪細(xì)胞的影響。 方法:以白藜蘆醇為原料,通過(guò)化學(xué)合成乙酰白藜蘆醇和甲基化白藜蘆醇,以核磁共振H譜、C譜、紫外光譜與質(zhì)譜對(duì)得到的產(chǎn)物進(jìn)行結(jié)構(gòu)解析;以高效液相色譜法測(cè)定含量和脂水分配系數(shù);細(xì)胞實(shí)驗(yàn)部分,實(shí)驗(yàn)分為空白對(duì)照組、白藜蘆醇對(duì)照組、甲基化白藜蘆醇組、乙酰白藜蘆醇組,用MTT法測(cè)定不同濃度的甲基化白藜蘆醇和乙酰白藜蘆醇對(duì)3T3-L1脂肪細(xì)胞增殖的抑制作用;甲基化白藜蘆醇、乙酰白藜蘆醇分別處理24h和48h后,采用AnnexinV-PI雙染法檢測(cè)定其誘導(dǎo)3T3-L1脂肪細(xì)胞細(xì)胞凋亡作用,采用RT-PCR法檢測(cè)定其對(duì)3T3-L1脂肪細(xì)胞中PPARγ的mRNA表達(dá)的影響。 結(jié)果:通過(guò)結(jié)構(gòu)確認(rèn),合成得到了乙酰白藜蘆醇和甲基化白藜蘆醇,,純度均大于99%;其脂水分配系數(shù)(25℃)分別為:LogP=1.49,4.20;細(xì)胞實(shí)驗(yàn)部分,與空白對(duì)照組比較,甲基化白藜蘆醇組、乙酰白藜蘆醇組均對(duì)3T3-L1脂肪細(xì)胞的細(xì)胞活力有明顯的抑制作用,且呈濃度時(shí)間依賴(lài)關(guān)系,其IC50分別為321.8103μg/mL,103.7696μg/mL(P㩳0.05);甲基化白藜蘆醇組、乙酰白藜蘆醇組藥物干預(yù)24h和48h后的3T3-L1脂肪細(xì)胞分布于Q2象限和Q4象限的比例逐漸增多,細(xì)胞凋亡率呈現(xiàn)上升趨勢(shì),表現(xiàn)出較為明顯的時(shí)間依賴(lài)關(guān)系(P㩳0.05);與空白對(duì)照組比較,甲基化白藜蘆醇組、乙酰白藜蘆醇組均下調(diào)3T3-L1脂肪細(xì)胞中的PPARγmRNA的表達(dá)(P㩳0.05)。 結(jié)論:乙酰白藜蘆醇和甲基化白藜蘆醇均有較好的脂溶性;甲基化白藜蘆醇和乙酰白藜蘆醇對(duì)3T3-L1脂肪細(xì)胞有抑制作用,可誘導(dǎo)3T3-L1脂肪細(xì)胞的凋亡,能下調(diào)3T3-L1脂肪細(xì)胞PPARγmRNA的表達(dá)水平。
[Abstract]:Objective: to synthesize the derivatives of resveratrol, acetyl resveratrol and methylresveratrol. Their effects on 3 T 3 L 1 adipocytes were observed. Methods: acetyl resveratrol and methylated resveratrol were chemically synthesized from resveratrol. The structures of the products were analyzed by NMR, UV and MS spectra. The cell experiment was divided into blank control group, resveratrol control group, methylated resveratrol group and acetyl resveratrol group. The inhibitory effects of methylated resveratrol and acetyl resveratrol on the proliferation of 3T3-L1 adipocytes were determined by MTT assay, and the effects of methylated resveratrol and acetyl resveratrol on the proliferation of 3T3-L1 adipocytes were observed after 24 h and 48 h treatment, respectively. The apoptosis of 3T3-L1 adipocytes was detected by Annexin V-PI double staining, and the expression of PPAR 緯 mRNA in 3T3-L1 adipocytes was detected by RT-PCR. Results: acetyl resveratrol and methylated resveratrol were synthesized by structure confirmation.
