【摘要】：目的通過觀察不同濃度叔丁基過氧化氫(t-BHP)作用不同時(shí)間后對人腎小球系膜細(xì)胞(HGMC)衰老指標(biāo)的影響,探討t-BHP在HGMC衰老過程中的作用。方法應(yīng)用10、30、50、100μmol/L t BHP刺激細(xì)胞,每天刺激1 h,連續(xù)5 d,刺激結(jié)束后48 h檢測細(xì)胞增殖情況、衰老相關(guān)β半乳糖苷酶(SAβ-gal)染色陽性細(xì)胞百分率、細(xì)胞周期變化及細(xì)胞形態(tài)改變確定t-BHP誘導(dǎo)HGMC衰老的最佳濃度。結(jié)果 30μmol/L t BHP刺激HGMC,每天1 h連續(xù)作用5次后可抑制HGMC增殖,且84%的HGMC SAβ-gal染色陽性,90%HGMC進(jìn)入G0~G1期;光鏡表現(xiàn)細(xì)胞體積增大、胞體扁平、胞質(zhì)顆粒增多;電鏡下細(xì)胞核膜內(nèi)陷,染色質(zhì)凝集、邊集,細(xì)胞質(zhì)空泡增多,出現(xiàn)髓樣溶酶體。結(jié)論 30μmol/L t BHP刺激HGMC后每天1 h連續(xù)作用5次,可作為誘導(dǎo)HGMC發(fā)生衰老的刺激因素。
[Abstract]:Objective to investigate the role of t-BHP in the aging of human glomerular Mesangial cells (HGMC) by observing the effects of different concentrations of t-BHPH on the aging of human glomerular Mesangial cells (HGMCs). Methods the cells were stimulated with 1030g / L 50 渭 mol / L t BHP for 1 hour for 5 days. After 48 hours of stimulation, the proliferation of cells and the percentage of positive cells with 尾 -galactosidase SA 尾 -galactosidase staining were detected. Cell cycle changes and cell morphology changes determined the optimal concentration of t-BHP induced HGMC senescence. Results HGMC stimulated by 30 渭 mol / L t BHP could inhibit the proliferation of HGMC after continuous treatment for 1 hour and 5 times a day, and 84% of HGMC SA 尾 -gal positive cells entered G0 / G 1 phase. Light microscopy showed that the cell volume was larger, the cell body was flat and the cytoplasmic granules were increased, and the nuclear membrane of HGMC was trapped under electron microscope. Chromatin agglutination, edge agglutination, cytoplasmic vacuoles increased and myeloid lysosomes appeared. Conclusion 30 渭 mol / L t BHP can be used as a stimulant to induce HGMC to senescence.
