【摘要】：脂蛋白相關(guān)磷脂酶A2(lipoprotein-associated Phospholipase A2,Lp-PLA2)是磷脂水解酶超家族中的一員,能水解低密度脂蛋白中的氧化修飾磷脂,產(chǎn)生溶血卵磷脂和氧化性游離脂肪酸兩種促炎介質(zhì)。最近研究發(fā)現(xiàn)Lp-PLA2能夠介導(dǎo)炎癥反應(yīng)以及促進(jìn)動(dòng)脈粥樣硬化(atherosclerosis,AS)的形成,現(xiàn)已成為一個(gè)新炎性反應(yīng)標(biāo)志物和心血管事件的獨(dú)立風(fēng)險(xiǎn)預(yù)示因子。針對這一動(dòng)脈粥樣硬化過程中的關(guān)鍵酶設(shè)計(jì)的抑制劑很多。其中Darapladib是目前最接近上市的一種Lp-PLA2特異性抑制劑,目前其Ⅲ期臨床試驗(yàn)已完成。Darapladib作為一種新興的抗AS分子靶點(diǎn)藥物,受到人們廣泛的關(guān)注。已有多項(xiàng)研究表明它能降低Lp-PLA2的活性,減輕炎癥反應(yīng),從而控制AS的發(fā)展與惡化。本文綜述了Lp-PLA2及其抑制劑Darapladib在AS中的作用機(jī)制以及近年來Darapladib的藥效學(xué)研究進(jìn)展。
[Abstract]:Lipoprotein related phospholipase A2 (lipoprotein-associated Phospholipase A2, Lp-PLA2), a member of the phosphatidic hydrolase superfamily, can hydrolyze oxidized phospholipids from low density lipoproteins and produce two kinds of proinflammatory mediators, hemolytic lecithin and oxidative free fatty acids. Recently, Lp-PLA2 has been found to mediate inflammatory reactions and promote activity. The formation of atherosclerosis (AS) has now become an independent risk predictor of a new inflammatory response marker and cardiovascular event. There are many inhibitors designed for the key enzymes in this atherosclerosis. Among them, Darapladib is one of the most closely related Lp-PLA2 specific inhibitors at present, and is currently in stage III The clinical trial has completed.Darapladib as a new anti AS molecular target drug, which has attracted wide attention. Many studies have shown that it can reduce the activity of Lp-PLA2, reduce the inflammatory reaction and control the development and deterioration of AS. This paper summarizes the mechanism of Lp-PLA2 and its inhibitor Darapladib in AS and the Dara in recent years. Progress in the pharmacodynamics of pladib.
