SPE及納米金涂層毛細管電泳檢測血漿中土霉素與多西環(huán)素

【摘要】:建立了混合型固相萃取及季銨化纖維素負載的納米金涂層毛細管電泳法快速檢測血漿中土霉素和多西環(huán)素的方法。采用季銨化纖維素負載的納米金復(fù)合材料(QC-Au NPs)對毛細管內(nèi)壁進行動態(tài)涂層,以抑制管壁對分析物的吸附,并對緩沖液p H值進行優(yōu)化。結(jié)果表明,該涂層能改善峰形和分離度,提高分離效率,其中土霉素的柱效提高了17.9倍。在涂層毛細管中,土霉素和多西環(huán)素的吸附被抑制,并在4 min內(nèi)出峰。在10~200μg/m L范圍內(nèi),土霉素和多西環(huán)素的峰面積與濃度的線性關(guān)系良好,相關(guān)系數(shù)(r)分別為0.997 6和0.995 2。血漿中土霉素和多西環(huán)素的加標回收率為88%~107%,相對標準偏差(RSD)為1.4%~7.7%。該方法快速、簡便,準確度和精密度高,適用于血漿中土霉素和多西環(huán)素的快速檢測。
[Abstract]:A method for rapid determination of oxytetracycline and doxycycline in plasma by capillary electrophoresis with nano-gold coated with mixed solid phase extraction and quaternary ammonium cellulose was established. Quaternary cellulose supported gold nanocomposite (QC-Au NPsNPs) was used to dynamically coat the inner wall of capillary to inhibit the adsorption of the analyte and optimize the pH value of buffer solution. The results showed that the coating could improve the peak shape, separation degree and separation efficiency, and the column efficiency of oxytetracycline was increased by 17.9 times. In the coated capillary, the adsorption of oxytetracycline and doxycycline was inhibited and the peak was obtained within 4 min. The linear relationship between the peak area and the concentration of oxytetracycline and doxycycline was good in the range of 10 渭 g / mL with correlation coefficients of 0.997 6 and 0.995 2, respectively. The recoveries of oxytetracycline and doxycycline in plasma were 88 and 107, and the relative standard deviation (RSD) was 1.4 and 7.7. The method is rapid, simple, accurate and accurate. It is suitable for the rapid determination of oxytetracycline and doxycycline in plasma.
