利福噴丁對大鼠免疫功能影響的研究

【摘要】:目的:了解應(yīng)用利福噴丁膠囊后能否增加或抑制與清除結(jié)核桿菌的免疫功能,并對免疫功能改變的可能機制進行初步研究;方法:1、取雄性SD大鼠70只,體重180±30g,隨機分為四組,分別為喂養(yǎng)前10只、喂養(yǎng)一個月、三個月,喂養(yǎng)三個月停喂一個月,后三組均20只,分為實驗組(10只)和對照組(10只)。2、喂養(yǎng)前10只自由飼養(yǎng),不做任何干預(yù),,另外三組實驗組用利福噴丁膠囊灌胃,對照組用生理鹽水灌胃,余喂養(yǎng)條件相同。3、用流式細(xì)胞術(shù)檢測CD_3~+、CD_4~+、CD_8~+及NK(CD-3CD_(161)~+)細(xì)胞計數(shù),用碳粒廓清法檢測巨噬細(xì)胞的吞噬功能。結(jié)果:1.實驗組與對照組各指標(biāo)比較中,喂養(yǎng)一月及喂養(yǎng)三月停喂一月時各指標(biāo)差異均無統(tǒng)計學(xué)意義,喂養(yǎng)3月實驗組與對照組各指標(biāo)比較中,兩組間的CD_3~+、CD_8~+、廓清指數(shù)、平均吞噬率四指標(biāo)之間的差異有統(tǒng)計學(xué)意義;兩組間的CD_4~+、CD_(161)~+、CD_4~+/CD_8~+三指標(biāo)差異無統(tǒng)計學(xué)意義.2.對照組各時間點與空白對照組比較中,兩組間CD-3~+、平均吞噬率、廓清指數(shù)之間的差異無統(tǒng)計學(xué)意義,CD_4~+、CD_8~+、CD_(161)~+、CD_4~+/CD_8~+在喂養(yǎng)一月、喂養(yǎng)三月,喂養(yǎng)三月停喂一月及空白組之間差異均有統(tǒng)計學(xué)意義;3.實驗組各時間點與空白對照組比較:CD_3~+、CD_8~+、CD_4~+/CD_8~+這三個指標(biāo)在喂養(yǎng)一月、喂養(yǎng)三月,喂養(yǎng)三月停喂一月及空白組之間差異均有統(tǒng)計學(xué)意義,CD_4~+、CD_(161)~+、平均吞噬率、廓清指數(shù)這4個指標(biāo)在各時間點間差異無統(tǒng)計學(xué)意義;結(jié)論:1.本實驗中,實驗組與對照組各指標(biāo)比較中,喂養(yǎng)一月及喂養(yǎng)三月停喂一月時各指標(biāo)差異均無統(tǒng)計學(xué)意義,表明喂養(yǎng)一月時利福噴丁膠囊的藥效未達到影響免疫功能的強度,喂養(yǎng)三月停喂一月,可能由于停藥后藥物在體內(nèi)的量減少或停藥后原有藥物隨體內(nèi)代謝清除,對各指標(biāo)的差異無顯著性的改變;2.喂養(yǎng)3月實驗組與對照組比較中,CD_3~+、CD_8~+、廓清指數(shù)、平均吞噬率4指標(biāo)差異有統(tǒng)計學(xué)意義,實驗組CD_3~+、CD_8~+、廓清指數(shù)、平均吞噬率的均數(shù)±標(biāo)準(zhǔn)差均比對照組增高,且各指標(biāo)差異有統(tǒng)計學(xué)意義,表明隨著喂藥時間的延長,藥物對個別免疫相關(guān)指標(biāo)有一定影響。3.對照組各時間點與空白對照組比較中,除CD_3~+、平均吞噬率、廓清指數(shù)外,指標(biāo)CD_4~+、CD_4~+/CD_8~+、CD_(161)~+、在喂養(yǎng)一月、喂養(yǎng)三月、喂養(yǎng)三月停喂一月及空白組之間差異均有統(tǒng)計學(xué)意義,該差異可能為樣本含量小導(dǎo)致的抽樣誤差或?qū)嶒灢僮髡`差所致;4.實驗組各時間點與空白對照組比較中:CD_3~+、CD_8~+、CD_4~+/CD_8~+這三個指標(biāo)在喂養(yǎng)一月、喂養(yǎng)三月、喂養(yǎng)三月及空白組之間差異均有統(tǒng)計學(xué)意義, 表明該藥對這些指標(biāo)的影響與藥物作用的時間呈正相關(guān)。CD_4~+、CD_(161)~+、平均吞噬率、廓清指數(shù)這4指標(biāo)在各時間點間差異無統(tǒng)計學(xué)意義,尚不能排除該藥對這些指標(biāo)無影響。
[Abstract]:Objective: to investigate whether the immune function of Mycobacterium tuberculosis can be increased, inhibited and eliminated by rifapentin capsule, and to study the possible mechanism of immune function change. Methods: 70 male SD rats, weighing 180 鹵30 g, were randomly divided into four groups. Ten rats were fed before, one month, three months, one month after feeding, 20 rats in the last three groups were divided into experimental group (n = 10) and control group (n = 10). 10 rats were fed freely without any intervention before feeding. The other three groups were fed with Rifopentin capsule, while the control group were fed with normal saline under the same feeding conditions. Flow cytometry was used to detect the count of CD3 ~ + CD4 ~ + CD4 ~ + and NKT CD-3CDD ~ (161 ~ +) cells, and the phagocytic function of macrophages was detected by carbon particle clearance method. The result is 1: 1. There was no significant difference in each index between the experimental group and the control group after feeding for one month and after feeding for three months. In the comparison of the indexes of the experimental group and the control group, the CD3 ~ + CD8 ~, and the clearance index between the two groups were not statistically significant. There was significant difference among the four indexes of average phagocytosis rate, and there was no significant difference in CD4 / CD4 / CD8 ~ 3 between the two groups. There was no significant difference in CD-3 ~, average phagocytosis rate and clearance index between the two groups at each time point compared with the blank control group. There was no statistical significance between the two groups. There were significant differences between the three months of feeding and the blank group. Compared with the blank control group, there were significant differences in the three indexes of CD4 ~ / CD8 ~ between the control group and the control group. The average phagocytosis rate was significantly higher than that in the control group after feeding for one month, three months after feeding for one month, and between the blank group and the control group, and the mean phagocytosis rate was higher than that in the control group, and the mean phagocytic rate was higher than that in the control group. There was no significant difference between the four indexes at different time points. Conclusion: 1. In this experiment, there was no significant difference in the indexes between the experimental group and the control group after feeding for one month and at the end of feeding for three months, which indicated that the efficacy of Rifopentin capsule did not reach the intensity of influence on immune function after feeding for one month. After feeding for three months or one month, there may be no significant difference between the two indexes because of the decrease in the amount of drug in the body after the withdrawal or the elimination of the original drug with the metabolism of the body after the withdrawal of the drug. Three months after feeding, there were significant differences in CD3 ~ + CD8 ~, clearance index and average phagocytosis rate between the experimental group and the control group. In the experimental group, CD3 ~ + CD8 ~, clearance index and mean 鹵standard deviation of the average phagocytosis rate were higher than those in the control group. The difference of each index was statistically significant, which indicated that with the prolongation of the feeding time, the drug had a certain influence on some immune-related indexes. In comparison between the control group and the blank control group at each time point, except for CD3, average phagocytosis rate and clearance index, the index CD4 ~ / CD4 ~ / CD8 ~ + CDT was significantly different between the control group and the control group. There were significant differences between the control group and the control group in feeding for one month, feeding for three months, feeding for one month, and the blank group. The difference may be caused by sampling error caused by small sample size or experimental operation error. Compared with the blank control group at each time point in the experimental group, there were statistically significant differences among the three indexes of the ratio of CD4 / CD8 to CD4 / CD8 ~ in the control group and the control group for the first month, the third month of feeding and the blank group. The results showed that the influence of the drug on these indexes was positively correlated with the time of the drug action. The average phagocytosis rate and clearance index of the four indexes were not statistically significant, and it could not be ruled out that the drug had no effect on these indexes.
