【摘要】：目的:探究我院自行配制咪達唑侖糖漿混合液在中國人群中的口感接受程度以及穩(wěn)定性。方法:咪達唑侖鹽酸注射液5mg/ml(M)與單糖漿850mg/l(S)按照不同體積配比混合制成5組不同濃度的咪達唑侖糖漿混合液(對照組5mg/mlM:S=1:0;A 組 3.3mg/mlM:S=1:0.5;B 組 2.5mg/mlM:S=1:1;C 組 2.0mg/mlM:S=1:1.5;D組1.67mg/mlM:S=1:2),采用滴定法將不同濃度咪達唑侖糖漿混合液按照隨機順序依次滴定于40名中國健康成人志愿者舌根部進行品嘗,志愿者每品嘗完一組立即漱口 60秒,并對該組咪達唑侖糖漿混合液口感選擇接受程度并打分,統(tǒng)計出接受程度最高的一組。配置接受程度最高組咪達唑侖糖漿混合液分別儲存于4℃和常溫下,采用質譜液相聯(lián)用法測量配置后第0,8,9,10,12,14,21天咪達唑侖糖漿混合液中咪達唑侖的藥物濃度,pH值測量儀測量咪達唑侖糖漿混合液pH值,肉眼觀察咪達唑侖糖漿混合液的顏色、澄明度及外觀的變化。結果:1、口感:咪達唑侖注射液與單糖漿體積配比為1:2(即咪達唑侖糖漿混合液濃度1.67mg/ml)時口感接受程度最高,接受程度評分顯著高于其他組,差異有統(tǒng)計學意義(P0.05)。2、穩(wěn)定性:1)咪達唑侖糖漿混合液21天內濃度穩(wěn)定性良好,常溫與4℃下濃度差異無統(tǒng)計學意義(P0.05)。2)咪達唑侖糖漿混合液21天內pH值呈線性增大趨勢,4℃相對于室溫pH值更穩(wěn)定,差異有統(tǒng)計學意義(P0.05),建議冷藏保存。3)咪達唑侖糖漿混合液21天內物理性狀穩(wěn)定性良好。結論:北京協(xié)和醫(yī)院自行配制的咪達唑侖糖漿混合液口感舒適,穩(wěn)定性良好,可作為一種良好的咪達唑侖注射液替代口服劑應用于中國臨床。
[Abstract]:Objective: to study the acceptability and stability of midazolam syrup mixture prepared by our hospital in Chinese population. Methods: midazolam hydrochloric acid injection (5mg / ml) was mixed with single syrup (850mg / L) to produce 5 groups of different concentrations of midazolam syrup (control group 5 mg / ml M: M: 1: 0A). The mixture of midazolam syrup was titrated in random order to taste the tongue root of 40 healthy Chinese adult volunteers. The volunteers gargled the mouthwash for 60 seconds immediately after each taste. The group with midazolam syrup was selected for acceptance and scored, and the group with the highest acceptance was counted.
