異綠原酸A在大鼠體內(nèi)的生物利用度和藥物代謝動力學(xué)

【摘要】:目的:研究異綠原酸A在大鼠體內(nèi)的生物利用度和藥代動力學(xué),為該制劑的臨床應(yīng)用提供參考。方法:建立大鼠血漿中異綠原酸A的HPLC檢測,檢測波長300 nm,流動相甲醇-0.1%磷酸水(50∶50)?疾齑笫蠼(jīng)靜脈注射(32 mg·kg-1)與灌胃(90 mg·kg-1)給予異綠原酸A后的血藥濃度變化,利用3P97軟件計算藥動學(xué)參數(shù),根據(jù)藥時曲線下面積AUC0-∞和給藥劑量,計算異綠原酸A的絕對生物利用度。結(jié)果:異綠原酸A在0.16~110.00 mg·L-1線性良好(R2=0.998);質(zhì)量濃度分別為0.43,6.88,55.00 mg·L-1的異綠原酸A的提取回收率分別為(89.43±2.84)%,(93.16±3.95)%,(85.91±2.04)%;日內(nèi)精密度RSD分別為11.8%,4.0%,4.0%,日間精密度RSD分別為6.5%,5.8%,5.8%。大鼠靜脈注射和灌胃異綠原酸A后,異綠原酸A在大鼠體內(nèi)的代謝過程均符合二室模型,消除半衰期分別為(29.49±0.75),(44.48±0.13)min,AUC0-∞分別為(355.40±32.58),(319.91±51.00)mg·min-1·L-1。異綠原酸A在大鼠體內(nèi)的絕對生物利用度30.71%。結(jié)論:異綠原酸A在大鼠體內(nèi)的過程符合線性動力學(xué)過程,且代謝快、半衰期短。
[Abstract]:Aim: to study the bioavailability and pharmacokinetics of Lv Yuan A in rats and to provide reference for its clinical application. Methods: HPLC detection of isophosphate A in rat plasma was established. The detection wavelength was 300nm.The mobile phase consisted of methanol-0.1% phosphoric acid water (50: 50).
