粘質(zhì)沙雷菌毒性噬菌體PS4的生物學(xué)特性及其在小鼠血清中的殺菌作用研究

【摘要】:目的研究粘質(zhì)沙雷菌毒性噬菌體PS4的生物學(xué)特性及其在小鼠血清中的抗菌活性。方法觀(guān)察PS4的噬菌斑及電鏡下形態(tài);測(cè)定PS4的噬菌譜、生長(zhǎng)曲線(xiàn)及其理化穩(wěn)定性,PS4在營(yíng)養(yǎng)肉湯培養(yǎng)基、新鮮小鼠血清和滅活小鼠血清中的殺菌作用,以及小鼠血清中影響PS4殺菌能力影響因素。結(jié)果 PS4為長(zhǎng)尾型噬菌體,在以粘質(zhì)沙雷菌臨床株S4為指示菌時(shí),PS4可形成直徑為1~1.5mm完全透明噬菌斑,其感染S4的潛伏期為10min,暴發(fā)量約為100PFU/cell,且S4對(duì)PS4的抗性突變率僅為10-7。PS4在pH 5~10的環(huán)境中具有較好的穩(wěn)定性,在50℃的環(huán)境中仍能長(zhǎng)時(shí)間保持活力。PS4在新鮮未滅活血清中對(duì)S4不具殺菌能力,但在56℃熱滅活30min的鼠血清中以及在營(yíng)養(yǎng)肉湯中的殺菌率均達(dá)99.99%,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 PS4具有一定的酸堿穩(wěn)定性和熱穩(wěn)定性,有望用于醫(yī)院環(huán)境"消毒",以減少多重耐藥粘質(zhì)沙雷菌所致的院內(nèi)感染的發(fā)生。但因其殺菌作用受血清中不耐熱成分的嚴(yán)重干擾,故不能用于治療由S4引起的小鼠全身感染。
[Abstract]:Objective to study the biological characteristics of bacteriophage PS4 and its antibacterial activity in mouse serum. Methods the plaque of PS4 and its morphology under electron microscope were observed, the bacteriophage spectrum, growth curve and physicochemical stability of PS4 were determined, and the bactericidal effects of PS4 in nutrient broth medium, fresh mouse serum and inactivated serum were determined. And the influence factors of PS4 bactericidal ability in serum of mice. Results PS4 was a long-tailed bacteriophage. When S4, a clinical strain of Shareh, was used as indicator, PS4 could form a completely transparent plaque with diameter of 1~1.5mm. The incubation period of S4 infection was 10 min, and the number of outbreaks was about 100 PFU / p cell. The mutation rate of S4 resistance to PS4 was only stable in pH 5 ~ 10 environment.
