【摘要】：新藥創(chuàng)制是復(fù)雜的智力活動(dòng),涉及科學(xué)研究、技術(shù)創(chuàng)造、產(chǎn)品開(kāi)發(fā)和醫(yī)療效果等多維科技活動(dòng)。每個(gè)藥物都有自身的研發(fā)軌跡,而構(gòu)建化學(xué)結(jié)構(gòu)是最重要的環(huán)節(jié),因?yàn)樗w了藥效、藥代、安全性和生物藥劑學(xué)等性質(zhì)。本欄目以藥物化學(xué)視角,對(duì)有代表性的藥物的成功構(gòu)建,加以剖析和解讀。在缺乏受體信息的情況下,以天然配體為出發(fā)點(diǎn),在結(jié)構(gòu)的變換和活性的變化中,逐漸演化出先導(dǎo)物,以及實(shí)施先導(dǎo)物的優(yōu)化。理性藥物設(shè)計(jì)不只限于計(jì)算機(jī)輔助的手段,構(gòu)效關(guān)系可以在多層次上進(jìn)行分析,化學(xué)結(jié)構(gòu)、構(gòu)象特征、功能基(藥效團(tuán))分布、物化性質(zhì)等,都可成為演繹和優(yōu)化結(jié)構(gòu)的依據(jù)。西咪替丁是理性藥物設(shè)計(jì)的另一種典范,充分應(yīng)用了藥物化學(xué)原理,如同系律、電子等排原理、拼合原理等。首創(chuàng)性藥物的可貴在于沒(méi)有借鑒和參考下開(kāi)辟了一個(gè)新的治療領(lǐng)域,包括靶標(biāo)的確證和藥物的成功。也由于首創(chuàng)藥物未能充分優(yōu)化,存在不足之處,為后續(xù)研發(fā)的替丁留下了空間,在模擬中后來(lái)居上。
[Abstract]:New drug creation is a complex intellectual activity involving multi-dimensional scientific and technological activities such as scientific research, technological creation, product development and medical effects. Each drug has its own R & D track, and the construction of chemical structures is the most important step because it covers pharmacodynamics, pharmacology, safety and biopharmaceutical properties. This column analyzes and interprets the successful construction of representative drugs from the perspective of drug chemistry. In the absence of receptor information, with the natural ligands as the starting point, in the transformation of the structure and the change of activity, the leader gradually evolved, and the optimization of the precursor was carried out. Rational drug design is not limited to computer-aided means. Structure-activity relationships can be analyzed at multiple levels, chemical structure, conformational characteristics, functional group (pharmacophore) distribution, physicochemical properties and so on, all of which can be used as the basis for deducing and optimizing the structure. Cimetidine is another model of rational drug design, which makes full use of the principles of pharmacochemistry, such as system law, electronic equal-arrangement principle, splicing principle and so on. The value of pioneering drugs lies in the opening up of a new field of treatment without reference, including the confirmation of targets and the success of drugs. Because the original drug is not fully optimized, there are some shortcomings, which leave room for the subsequent research and development of tetidine, and become better in the simulation.
