【摘要】：以添加鎮(zhèn)靜催眠候選新藥N6-羥芐腺嘌呤核苷[N6-(4-hydroxybenzyl)adenine riboside,NHBA]的組織勻漿切片作為考察對(duì)象,對(duì)空氣動(dòng)力輔助離子化質(zhì)譜成像系統(tǒng)(Air Flow Assisted Ionization Mass Spectrometry Imaging,AFAI-MSI)的關(guān)鍵參數(shù)進(jìn)行考察與優(yōu)化,以保證最佳條件進(jìn)行樣品檢測。在此基礎(chǔ)上,對(duì)預(yù)處理的整體大鼠組織切片進(jìn)行平行的連續(xù)兩次質(zhì)譜成像分析,考察了其內(nèi)源性代謝物在成像分析過程中是否發(fā)生變化。通過對(duì)采集數(shù)據(jù)中關(guān)鍵質(zhì)譜峰的篩選等處理步驟,并采用可視化的主成分分析(PCA)方法,開展了組織樣品內(nèi)源性代謝物的穩(wěn)定性分析,最終驗(yàn)證了采用該樣品前處理和質(zhì)譜成像方法,能夠保證組織切片樣品中內(nèi)源性代謝物的穩(wěn)定性,為質(zhì)譜成像分析結(jié)果提供了可靠依據(jù)。
[Abstract]:The key parameters of air Flow Assisted Ionization Mass Spectrometry imaging system (AFAI-MSI) were optimized by using tissue homogenate sections supplemented with N6-(4-hydroxybenzyl)adenine riboside (NHBA), a sedative hypnotic candidate drug. To ensure the best conditions for sample detection. On this basis, the pretreated tissue sections of the whole rat were analyzed by two consecutive mass spectrometry images, and the changes of endogenous metabolites in the process of imaging analysis were investigated. The stability analysis of endogenous metabolites in tissue samples was carried out by screening the key mass spectrum peaks and using the visual principal component analysis (PCA) method. Finally, it is proved that the method of sample pretreatment and mass spectrometry imaging can ensure the stability of endogenous metabolites in tissue slice samples, which provides a reliable basis for the analysis results of mass spectrometry imaging.
