【摘要】：目的:綜述腺相關(guān)病毒(AAV)載體的分離純化方法,為AAV的規(guī)�；苽涮峁﹨⒖�。方法:以"AAV""Purity""Chromatography""腺相關(guān)病毒""純化"等為關(guān)鍵詞,組合檢索2000年1月-2015年8月在Pub Med、Web of Science、Elsevier、中國(guó)知網(wǎng)、維普等數(shù)據(jù)庫中有關(guān)AAV分離純化的文獻(xiàn),選擇有代表性的文獻(xiàn)進(jìn)行歸納、總結(jié),分別從AAV純化的超速離心法、液相色譜法、化學(xué)試劑法和超濾法等4個(gè)方面進(jìn)行綜述。結(jié)果:共查閱到相關(guān)文獻(xiàn)260余篇,其中有效文獻(xiàn)48篇。液相色譜法純化AAV表現(xiàn)優(yōu)異,親和色譜、離子交換色譜等色譜技術(shù)已經(jīng)廣泛用于AAV1、2、4、5、6、8、9等不同血清型載體的分離純化,甚至較難分離的AAV的空殼載體也可以實(shí)現(xiàn)分離。但是每種血清型載體的理化性質(zhì)不盡相同,很難用同一種方法純化所有載體。結(jié)論:液相色譜法在規(guī)模化純化AAV載體具備良好優(yōu)勢(shì),但是應(yīng)該根據(jù)不同血清型載體理化特征建立對(duì)應(yīng)的純化工藝。
[Abstract]:Aim: to summarize the methods of isolation and purification of adeno-associated virus (AV) vector and to provide reference for the preparation of AAV on a large scale. Methods: using "AAV", "Purity," Chromatography "," adeno-associated virus "and" purification "as the key words, the literature on the separation and purification of AAV from Pub MedAAV Web of Science Elsevier, China knowledge Network, Weip and other databases from January 2000 to August 2015 was searched, and the representative literature was selected to summarize and summarize. In this paper, the purification of AAV by ultracentrifugation, liquid chromatography, chemical reagent and ultrafiltration were reviewed. Results: more than 260 articles were reviewed, 48 of which were effective. The purification of AAV by liquid chromatography is excellent. Affinity chromatography, ion exchange chromatography and other chromatographic techniques have been widely used in the separation and purification of different serotypes, such as AAV1 / AV _ (1), ~ (2 +) ~ (2 +), ~ (-) ~ (5) ~ (-) and so on. Even the empty shell carrier of AAV, which is more difficult to separate, can be separated. However, the physical and chemical properties of each serotype vector are different, so it is difficult to purify all the vectors by the same method. Conclusion: liquid chromatography has a good advantage in the large-scale purification of AAV vectors, but the corresponding purification process should be established according to the physicochemical characteristics of different serotype carriers.
