赭曲霉轉(zhuǎn)化洋地黃毒苷的工藝優(yōu)化和產(chǎn)物活性分析

【摘要】:以赭曲霉Rn405轉(zhuǎn)化洋地黃毒苷的微生物轉(zhuǎn)化工藝為研究對象,對助溶劑種類、底物濃度、金屬離子種類和濃度等關(guān)鍵參數(shù)進(jìn)行優(yōu)化,確定的最優(yōu)轉(zhuǎn)化條件為:發(fā)酵培養(yǎng)基中添加1 mmol/L Mn2+離子,28℃,180 rpm振蕩培養(yǎng)28 h時(shí)加入溶解于DMSO的底物溶液,使其終濃度為100 mg/L,然后在相同的條件下轉(zhuǎn)化96 h。此時(shí),底物的轉(zhuǎn)化率和11α-羥基洋地黃毒苷元的生成率分別達(dá)到了100%和25.4%,比優(yōu)化前分別提高了33.1%和14.5%。通過斯氏灌流法和Langendorff灌流法,探討兩個(gè)轉(zhuǎn)化產(chǎn)物對蟾蜍和大鼠心肌細(xì)胞收縮能力的影響。結(jié)果表明,洋地黃毒苷元對心肌細(xì)胞收縮率的影響和底物洋地黃毒苷相當(dāng),而11α-羥基洋地黃毒苷元對心肌細(xì)胞收縮率的影響比底物洋地黃毒苷和洋地黃毒苷元均有所增強(qiáng)。
