【摘要】：目的:建立同時(shí)測定人血漿中丙戊酸鈉(VPA)及其代謝產(chǎn)物2-丙基-2-戊烯酸(2-ene-VPA)濃度的方法。方法:血漿樣品經(jīng)環(huán)己烷沉淀蛋白后,用2,4'-二溴苯乙酮進(jìn)行衍生化,以正辛酸為內(nèi)標(biāo),采用反相高效液相色譜法測定。色譜柱為Zorbax SB-C_(18),流動相為乙腈-水(65:35,V/V),流速為1 mL/min,柱溫為35℃,紫外檢測波長為258 nm,進(jìn)樣量為20μL。結(jié)果:VPA、2-ene-VPA血藥濃度分別在5.0~200.0、0.5~20.0μg/mL范圍內(nèi)線性關(guān)系良好(r均為0.999 9,n=5),定量下限分別為5.0、0.5μg/mL;日內(nèi)、日間RSD均小于5%,方法回收率分別為95.99%~98.80%、97.40%~98.17%,提取回收率分別為80.46%~86.23%、80.45%~85.61%。采用該法測得10例癲癇患兒VPA血藥濃度為27.4~93.2μg/mL,2-ene-VPA血藥濃度為0.85~3.94μg/mL。結(jié)論:該方法操作簡便、專屬性強(qiáng),可用于VPA血藥濃度檢測及藥動學(xué)研究。
[Abstract]:Aim: to establish a method for simultaneous determination of valproate sodium valproate and its metabolite 2-propyl-2-pentenene-VPA in human plasma.
