透明質(zhì)酸修飾的尿酸酶多囊脂質(zhì)體的體外特性及藥效學(xué)過程

【摘要】:目的研究透明質(zhì)酸修飾的尿酸酶(UC)多囊脂質(zhì)體(uricase in hyaluronic acid-uricase multivesicular liposomes,UHMVLs)的體外特性及其在大鼠體內(nèi)的藥效學(xué)。方法采用復(fù)乳法制備UHMVLs并測(cè)定包封率及理化特性。取12只健康雄性SD大鼠,模型組采用次黃嘌呤和氧嗪酸鉀建立高尿酸血癥大鼠模型(n=3),UHMVLs組(n=3)和UC組(n=3)分別于建模后尾靜脈注射UHMVLs和游離UC,并以正常組(n=3)為對(duì)照,測(cè)定大鼠血清中尿酸水平。結(jié)果 UHMVLs的平均包封率為(62.48±3.87)%(n=3)。UHMVLs和UC最適溫度均為40℃,最適pH值分別為8.0和8.5。在同一溫度(20~70℃)和pH(6.5~9.5)條件下,UHMVLs中UC的活性均高于游離UC(P0.05)。除1、36、48h外,其余各時(shí)間點(diǎn)UHMVLs降低高尿酸血癥大鼠模型血清中尿酸水平的效果均較游離UC更顯著(P0.05)。結(jié)論在相同條件下,UHMVLs不僅能提高UC的活性,還可以增強(qiáng)UC的穩(wěn)定性;UHMVLs在大鼠體內(nèi)降低血尿酸水平的能力優(yōu)于游離UC。
[Abstract]:Aim to study the in vitro characteristics of hyaluronic acid-modified uricase in hyaluronic acid-uricase multivesicular liposome UHMVLs and its pharmacodynamics in rats. Methods UHMVLs was prepared by double emulsion method and the encapsulation efficiency and physicochemical properties were determined. Twelve healthy male SD rats were treated with hypoxanthine and potassium oxazinate to establish hyperuricemia rat models of hyperuricemia. (1) UHMVLs and free UC3 were injected into tail vein of model rats, respectively, and the control group was treated with UHMVLs (UHMVLs group) and UHMVLs group (UHMVLs group) (UHMVLs group) and UHMVLs group (UHMVLs group). The level of uric acid in serum of rats was determined. Results the average entrapment efficiency of UHMVLs was 62.48 鹵3.87)%(n=3).UHMVLs and the optimum temperature of UC was 40 鈩,
