四苯乙烯類熒光探針的合成及在生化檢測(cè)領(lǐng)域的應(yīng)用

【摘要】:對(duì)于傳統(tǒng)的熒光化合物,它們?cè)谌芤籂顟B(tài)下,會(huì)發(fā)出很強(qiáng)的熒光,但在聚集態(tài)或者固態(tài)時(shí)熒光性能減弱甚至消失,,即產(chǎn)生熒光淬滅效應(yīng)(Aggregation-causedquench,ACQ)。但人們?nèi)粘I钪校瑐鞲衅鞯鹊膽?yīng)用,往往需要制成一些固體、薄膜材料或者其他的一些材料,才能方便的應(yīng)用。這種ACQ現(xiàn)象的存在使得這類材料在傳感器等的應(yīng)用中受到了很大的限制。 在2001年,唐本忠等人發(fā)現(xiàn)了硅雜環(huán)戊二烯(siloles)分子具有相反的熒光性能。他們完全溶解在溶液中時(shí),熒光信號(hào)微弱,當(dāng)其聚集或形成固態(tài)時(shí),熒光信號(hào)顯著增強(qiáng)。他們將這一現(xiàn)象命名為聚集誘導(dǎo)發(fā)光(Aggregation-induced emission,AIE)現(xiàn)象。AIE化合物這一性質(zhì)使其可以作為探針?lè)肿釉诜肿、離子檢測(cè)領(lǐng)域以及生物檢測(cè)領(lǐng)域得到廣泛應(yīng)用。 本文以具有AIE性能的四苯乙烯類化合物為研究對(duì)象,分別合成了四苯乙烯、帶負(fù)電荷和正電荷的四苯乙烯分子,考察了三種化合物在分子、離子檢測(cè)領(lǐng)域的應(yīng)用。應(yīng)用于表面活性劑臨界膠束濃度(Critical micelle concentration,CMC)、重金屬離子以及植酸分子的檢測(cè)。 合成了四苯乙烯,利用其聚集誘導(dǎo)熒光性能,對(duì)三種不同類型的表面活性劑吐溫-80、十二烷基硫酸鈉和十六烷甲基溴化銨的CMC進(jìn)行了測(cè)定,并與參考文獻(xiàn)中的數(shù)據(jù)做了對(duì)照。 合成了帶有負(fù)電荷的四苯乙烯磺酸鈉鹽,在水相體系中對(duì)一系列的金屬離子進(jìn)行了檢測(cè),發(fā)現(xiàn)其對(duì)重金屬鉛離子有特異性識(shí)別作用。在此基礎(chǔ)上,將該方法用于中藥提取液中鉛離子的檢測(cè),結(jié)果證明同樣有選擇識(shí)別作用。 合成了帶正電荷的四苯乙烯季銨鹽,利用六個(gè)磷酸根離子的植酸分子誘導(dǎo)帶正電荷的四苯乙烯季銨鹽分子聚集,實(shí)現(xiàn)了對(duì)植酸和植酸酶的檢測(cè)。
[Abstract]:For the traditional fluorescent compounds, they emit very strong fluorescence in the solution state, but the fluorescence performance weakens or even disappears in the aggregate state or the solid state, that is, the fluorescence quenching effect Aggregation-causedquench (ACQ) is produced. However, in daily life, sensors and other applications often need to be made into some solids, thin film materials or other materials, in order to facilitate the application. Due to the existence of this ACQ phenomenon, the application of this kind of material in sensors is greatly restricted. In 2001, Tang Benzhong et al found that silyl heterocyclopentadiene siloles molecules have the opposite fluorescence properties. When they are completely dissolved in the solution, the fluorescence signal is weak, and when it aggregates or forms a solid state, the fluorescence signal increases significantly. They named this phenomenon as agglomeration induced emission AIE.AIE compounds can be widely used as probe molecules in molecular, ion detection and biological detection fields. In this paper, tetrastyrene compounds with AIE properties were synthesized, tetrastyrene molecules with negative charge and positive charge were synthesized, and the applications of three compounds in the field of molecular and ion detection were investigated. It has been applied to the detection of critical micelle concentration, heavy metal ions and phytic acid molecules. Tetrastyrene was synthesized and its aggregation induced fluorescence property was used to determine the CMC of three kinds of surfactants Tween-80, sodium dodecyl sulfate and cetylmethylammonium bromide. Sodium tetrastyrene sulfonate with negative charge was synthesized and a series of metal ions were detected in aqueous system. On this basis, the method has been applied to the determination of lead ion in the extract of traditional Chinese medicine. The results show that the method also has selective recognition effect. Tetrastyrene quaternary ammonium salt with positive charge was synthesized. Phytic acid molecules with six phosphate ions were used to induce the aggregation of positively charged tetrastyrene quaternary ammonium salts. The detection of phytic acid and phytase was realized.
