【摘要】：目的觀察不同濃度前列腺素E2(PGE2)刺激對脂多糖(LPS)誘導(dǎo)小鼠骨髓源性樹突細(xì)胞(DCs)成熟及EP4受體表達(dá)的影響。方法采用重組小鼠粒細(xì)胞-巨噬細(xì)胞集落刺激因子(rm GM-CSF)、重組小鼠白細(xì)胞介素4(rm IL-4)刺激小鼠骨髓源細(xì)胞,誘導(dǎo)生成DCs;經(jīng)LPS(100 ng/ml)誘導(dǎo)成熟后,用不同濃度PGE2(0.625、1.25、2.5、5、10、20nmol/L)刺激DCs 24 h,流式細(xì)胞術(shù)檢測DCs表型CD40、CD83、MHC-Ⅱ的表達(dá)和抗原攝取功能,熒光間標(biāo)法檢測小鼠骨髓源DCs細(xì)胞膜上EP4的表達(dá),以平均熒光強(qiáng)度表示表達(dá)的高低;MTT法檢測經(jīng)PGE2及LPS誘導(dǎo)成熟的DCs對T細(xì)胞增殖的作用。結(jié)果流式細(xì)胞術(shù)結(jié)果顯示PGE2(2.5、5、10 nmol/L)能明顯上調(diào)CD40、CD83、MHC-Ⅱ的表達(dá);PGE2(1.25、2.5、5、10、20 nmol/L)能明顯抑制DCs的抗原攝取功能;MTT法檢測結(jié)果顯示PGE2(2.5、5、10 nmol/L)刺激DCs能促進(jìn)T細(xì)胞增殖的作用;熒光間標(biāo)法檢測結(jié)果顯示PGE2(2.5、5、10 nmol/L)明顯升高DCs細(xì)胞膜上EP4表達(dá)。結(jié)論 PGE2可以調(diào)節(jié)DCs功能,該作用可能與其調(diào)節(jié)EP4受體表達(dá)相關(guān)。
