【摘要】：大多數(shù)人類基因組(98%)不編碼蛋白質(zhì),但是大部分的DNA序列(約75%)能夠轉(zhuǎn)錄為非編碼RNA(Non-coding RNA,ncRNA)。并且,近年來的研究發(fā)現(xiàn),長鏈非編碼RNA(Long Non-Coding RNA,lncRNA,指轉(zhuǎn)錄長度大于200kb的非編碼RNA)在高等生物的許多重要生物過程中發(fā)揮作用。它們通常通過與多種功能性基因的相互調(diào)控來發(fā)揮作用,同時(shí),它們的異�？稍斐砂┌Y等多種疾病的形成。其中,極長基因間非編碼RNA(Very Long Integenic Non-Coding RNAs,vlincRNAs)為一類特殊的長鏈非編碼RNA,目前的研究結(jié)果已經(jīng)顯現(xiàn)了其參與到早期胚胎發(fā)育以及癌癥發(fā)生的過程中,但仍有潛在功能未被證實(shí)。vlincRNAs為癌癥發(fā)生的早期階段的研究提供了一個(gè)新方向�？紤]到大量未被發(fā)現(xiàn)的lnc-和vlincRNAs以及它們在細(xì)胞各項(xiàng)生理活動中的作用,這些轉(zhuǎn)錄物或許掌握著一個(gè)細(xì)胞發(fā)育或癌變的關(guān)鍵。并且,越來越多的研究表明,lncRNAs在抗癌藥物的作用過程中發(fā)揮著重要作用。因此,這些轉(zhuǎn)錄產(chǎn)物可能與很多抗癌藥物作用的分子機(jī)制相關(guān)。本論文通過研究抗癌藥物對vlincRNAs表達(dá)的影響,從而了解vlincRNAs的功能。具體地說,我們利用單分子測序平臺,對使用多種抗癌藥物處理后的人類白血病細(xì)胞K562進(jìn)行RNA測序,對測序結(jié)果進(jìn)行分析,篩選vlincRNAs。本文特別關(guān)注兩種抗癌藥物——伊馬替尼(Gleevec)和司美替尼。其中,伊馬替尼是一種公認(rèn)的治療作用較好且銷量很高的抗癌藥物,司美替尼是一個(gè)仍處于實(shí)驗(yàn)階段的抗癌藥物。我發(fā)現(xiàn)經(jīng)過這兩種藥物處理K562細(xì)胞系之后,一些vlincRNAs的表達(dá)發(fā)生了顯著的變化。另外,我通過多次重復(fù)實(shí)驗(yàn)以及多種實(shí)驗(yàn)手段已經(jīng)確定了KVUG1、KVUG2和KVUG3這3條vlincRNAs可能參與到了伊馬替尼所調(diào)控的細(xì)胞增殖、新陳代謝和細(xì)胞免疫過程中。
[Abstract]:Most human genomes do not encode proteins, but most of the DNA sequences can be transcribed into non-coding RNA(Non-coding RNAs. Moreover, in recent years, it has been found that long chain noncoding RNA(Long Non-Coding (LNNRNAs), which refer to non-coding RNAs whose transcription length is longer than 200kb, plays an important role in many important biological processes in higher organisms. They usually function through mutual regulation with multiple functional genes, and their abnormalities can lead to the formation of many diseases, such as cancer. Among them, the extremely long intergenic noncoding RNA(Very Long Integenic Non-Coding RNAsvlincRNAs1 is a special kind of long chain noncoding RNAs. The present research results have shown that it is involved in the process of early embryonic development and carcinogenesis. But there are still potential functions that have not been confirmed. VlincRNAs provide a new direction for the early stage of cancer development. Given the large number of undiscovered LNC-and vlincRNAs and their role in various cellular physiological activities these transcripts may hold the key to cell development or carcinogenesis. Moreover, more and more studies have shown that lncRNAs play an important role in the action of anticancer drugs. Therefore, these transcription products may be related to the molecular mechanism of many anticancer drugs. In order to understand the function of vlincRNAs, we studied the effect of anticancer drugs on the expression of vlincRNAs. Specifically, we sequenced the RNA of human leukemia cell K562 treated with various anticancer drugs using single molecular sequencing platform, and analyzed the sequencing results to screen vlinc RNAs. This paper focuses on two anticancer drugs-imatinide Gleevec) and smectini. Among them, imatinib is recognized as a good therapeutic and high-volume anticancer drug, and smectini is still in the experimental stage of anticancer drugs. I found that there were significant changes in the expression of some vlincRNAs in K562 cells treated with these two drugs. In addition, I have confirmed that KVUG1KVUG2 and KVUG3 may be involved in the cell proliferation, metabolism and cellular immunity regulated by imatinib.
