一株銀杏內(nèi)生真菌抗菌代謝產(chǎn)物的分離與鑒定

【摘要】:目的分離并鑒定銀杏內(nèi)生真菌產(chǎn)生的抗菌活性物質(zhì)。方法在一株鐮刀菌屬內(nèi)生真菌的發(fā)酵液中檢測(cè)出了較強(qiáng)的抗革蘭陽(yáng)性菌的活性,抗菌活性篩選的檢定菌為短小芽孢桿菌[CMCC(B)63202]。發(fā)酵液經(jīng)液液萃取、硅膠柱層析、高效液相分離得到化合物A和B。液相色譜條件:以Shim-pack C_(18)(250mm×10mm,5μm)為固定相,以甲醇-水(5:6)為流動(dòng)相,流速1.0m L/min,檢測(cè)波長(zhǎng)254nm,進(jìn)樣量20μL。結(jié)果經(jīng)結(jié)構(gòu)解析,化合物A的分子量為639,由3個(gè)α-羥基-β-甲基-丁酸(Hiv)與3個(gè)N甲基纈氨酸(MeVal)相互間隔地縮合成一個(gè)環(huán)狀對(duì)稱(chēng)結(jié)構(gòu),與文獻(xiàn)中恩鐮孢菌素B的結(jié)構(gòu)一致;衔顱的分子量為653,與恩鐮孢菌素B相差一個(gè)CH_2,推測(cè)與恩鐮孢菌素B_1或恩鐮孢菌素D同質(zhì)。結(jié)論該銀杏內(nèi)生真菌產(chǎn)生的抗菌活性物質(zhì)為恩鐮孢菌素。
[Abstract]:Objective to isolate and identify the antimicrobial active substances produced by endophytic fungi of Ginkgo biloba. Methods A strain of Fusarium endophytic fungi was used to detect the activity of Gram-positive bacteria in the fermentation broth of Fusarium. The antimicrobial activity was screened by Bacillus pumilus [CMCC(B)63202]. Compounds A and B were separated from fermentation broth by liquid-liquid extraction, silica gel column chromatography and high performance liquid chromatography. The liquid chromatographic conditions were as follows: Shim-pack C_(18)(250mm 脳 10mm-1 5 渭 m) as stationary phase, methanol-water 5: 6) as mobile phase, flow rate 1.0 mL / min, detection wavelength 254 nm, sample volume 20 渭 L. Results the molecular weight of compound A was 639. Three 偽 -hydroxy- 尾 -methyl-butyric acid (Hiv) and three N-methyl valine (MeVala) were separated to form a cyclic symmetric structure, which was consistent with the structure of Fusarium B in the literature. The molecular weight of compound B is 653, which is one CHS _ 2 difference from that of Fusarium B, which is conjectured to be the same as Fusarium B _ 1 or E _ S _ D. Conclusion the antimicrobial activity produced by endophytic fungi of Ginkgo biloba is Fusarium mongolicum.
