乙醇暴露對(duì)小鼠肝臟脂質(zhì)合成的影響

【摘要】:目的觀察小鼠不同時(shí)間乙醇暴露,其肝臟脂質(zhì)合成情況。方法 8周齡雄性ICR小鼠隨機(jī)分為空白對(duì)照組和乙醇實(shí)驗(yàn)組,實(shí)驗(yàn)組小鼠隨機(jī)分為單次和連續(xù)4 d兩個(gè)不同的時(shí)間組,均給予50%(V/V)乙醇溶液(4 g/kg體重)灌胃,對(duì)照組給予等體積的生理鹽水。所有小鼠均于末次灌胃12 h后剖殺,取血清用于檢測(cè)血液生化指標(biāo);取肝臟并稱重,計(jì)算肝臟系數(shù),檢測(cè)甘油三酯(triglycerides,TG)和總膽固醇(total cholesterol,TCH)含量,蘇木素伊紅(hematoxylin and eosin,HE)染色觀察其病理學(xué)改變,油紅O染色觀察其脂質(zhì)沉積;反轉(zhuǎn)錄酶-聚合酶鏈鎖反應(yīng)(reverse transcription-polymerase chain reaction,RT-PCR)檢測(cè)乙酰輔酶A羧化酶(acetyl coenzyme carboxylase,ACC)、脂肪酸合成酶(fatty acid synthase,FAS)和硬脂酰輔酶A去飽和酶-1(stearoyl coenzyme A desaturase-1,SCD-1)的mRNA水平。結(jié)果與對(duì)照組相比,單次或4 d乙醇暴露小鼠肝臟系數(shù)均增加(均有P0.05);血清和肝臟TG含量升高(均有P0.05);HE染色和油紅O染色顯示,肝臟發(fā)生脂質(zhì)沉積;FAS、ACC和SCD-1 mRNA水平上調(diào)(均有P0.05)。結(jié)論單次或連續(xù)4 d乙醇暴露均可導(dǎo)致肝臟脂質(zhì)沉積,并通過(guò)增加小鼠肝臟TG合成來(lái)完成。
[Abstract]:Objective to observe the lipid synthesis in liver of mice exposed to ethanol at different time. Methods 8-week-old male ICR mice were randomly divided into two groups: blank control group and ethanol experimental group. The experimental group mice were randomly divided into two groups: the control group and the control group. The mice in the experimental group were given 50 V / V ethanol solution for 4 g/kg. The control group was given saline of the same volume. All mice were dissected 12 hours after the last gavage, and the serum was taken for the determination of biochemical indexes in blood, the liver was taken and weighed, the liver coefficient was calculated, the contents of triglycerides triglycerides TGG) and total cholesterol total cholesterol total cholesterol (TC) were measured. The pathological changes were observed by hematoxylin and eosin hehe staining and lipid deposition was observed by oil red O staining. Reverse transcription-polymerase chain reactionation (RT-PCR) was used to detect the mRNA levels of acetyl coenzyme carboxylase1, fatty acid synthase fatty acid synthaseFASand stearoyl coenzyme A desaturase-1SCD-1. Results compared with the control group, the liver coefficients of mice exposed to ethanol at single or 4 days were all increased (P 0.05), and the serum and liver TG levels were increased (both P 0.05 he staining and oil red O staining showed that liver lipid deposition FASA ACC and SCD-1 mRNA levels were up-regulated (both P 0.05). Conclusion single or continuous ethanol exposure for 4 days can lead to lipids deposition in the liver, which can be accomplished by increasing the synthesis of TG in the liver of mice.
