納米銀對皮膚細胞半通道活性的影響及其在細胞增殖抑制中的作用

【摘要】:目的:探討納米銀(silver nanoparticles,AgNPs)對皮膚細胞半通道活性的影響,并進一步研究半通道活性的改變在AgNPs誘導的細胞增殖抑制中的作用。方法:不同濃度(0、0.1、0.2、1.0、5.0、10.0、15.0和20.0μg/cm~2)的AgNPs處理人永生化表皮細胞系(HaCaT)24 h,采用碘化丙啶(propidium iodide,PI)染料排除法測定細胞活性,采用溴化乙錠(ethidium bromide,EB)攝取法測定細胞半通道活性。通過以常規(guī)密度(4×10~4個/cm~2)或低密度(1×10~4個/cm~2)接種細胞及采用半通道抑制劑甘珀酸(carbenoxolone,CBX)預處理的方法調(diào)節(jié)細胞半通道活性,采用細胞增殖-毒性檢測試劑盒(cell counting kit-8,CCK-8)檢測半通道活性受抑制、不同濃度AgNPs作用后的HaCaT細胞的增殖水平。結(jié)果:當AgNPs濃度低于10μg/cm~2時,HaCaT細胞的細胞活性未受明顯影響。10μg/cm~2的AgNPs作用2、12和24 h可導致細胞半通道活性升高至對照組的116.67%、124.85%和139.53%,與對照組相比差異均具有統(tǒng)計學意義(P0.01),且25、50和100μmol/L CBX預處理可明顯抑制10μg/cm~2AgNPs作用24 h后的細胞的半通道活性(P0.01)。此外,100μmol/L CBX預處理可升高低密度和常規(guī)密度下10μg/cm~2AgNPs作用24 h后細胞的增殖水平,其中,低密度下,CBX預處理組與AgNPs單獨處理組相比,細胞增殖水平顯著升高(P0.01)。結(jié)論:一定濃度的AgNPs可激活HaCaT細胞的半通道,半通道活性升高在AgNPs引發(fā)的細胞增殖抑制效應中具有重要作用。
[Abstract]:Aim: to investigate the effects of silver silver nanoparticlesclines (Ag NPs) on the half-channel activity of skin cells, and to investigate the role of the changes of half-channel activity in the inhibition of cell proliferation induced by AgNPs. Methods: human immortalized epidermal cell line HaCaT was treated with AgNPs at different concentrations of 0.1g / cm ~ (2) and 20.0 渭 g / cm ~ (-2) respectively. The cell activity was determined by propidium iodide PII dye exclusion method, and the half-channel activity was measured by ethidium bromidee uptake method. Cells were inoculated with conventional density of 4 脳 10 ~ 4 / cm ~ (-2) or low density of 1 脳 10 ~ (4) / cm ~ (-2) and treated with carbenoxolone (CBX), a half-channel inhibitor, to regulate the cell half-channel activity. Cell counting kit-8 CCK-8 was used to detect the inhibition of half-channel activity and the proliferation level of HaCaT cells treated with different concentrations of AgNPs. Results: when the concentration of AgNPs was lower than 10 渭 g / cm ~ 2, the cell activity of HaCaT cells was not significantly affected by AgNPs of 10 渭 g/cm~2 for 2 h and 24 h, and the half-channel activity increased to 116.67 ~ 124.85% and 139.53% of the control group. The difference was statistically significant compared with the control group. Pretreatment with 25 渭 mol/L and 100 渭 mol/L CBX significantly inhibited the half-channel activity of P0.01G after 24 h treatment with 10 渭 g/cm~2AgNPs. In addition, preconditioning with 100 渭 mol/L CBX could increase the proliferation level of cells treated with 10 渭 g/cm~2AgNPs at low density and normal density for 24 h, and the proliferation level of cells in low density pretreated group was significantly higher than that in AgNPs alone group. Conclusion: a certain concentration of AgNPs can activate the half-channel of HaCaT cells, and the increase of half-channel activity plays an important role in the inhibition of cell proliferation induced by AgNPs.
