【摘要】：結(jié)合預(yù)試驗結(jié)果,首先確定前列地爾微乳的處方組成,再通過星點設(shè)計-效應(yīng)面法和繪制電導(dǎo)率-含水量曲線,確定了前列地爾微乳處方中各組分的優(yōu)化用量;最后以粒徑和藥物含量為指標(biāo),優(yōu)化了微乳制備時的攪拌速度、時間和溫度等工藝參數(shù)。結(jié)果表明,前列地爾微乳優(yōu)化處方為:Solutol HS 15 8%、蛋黃卵磷脂2%、中鏈甘油三酸酯4%、甘油6%和水80%;制備時攪拌的速度、溫度和時間分別為300 r/min、30℃和0.5 h。照上述優(yōu)化處方制備的3批前列地爾微乳粒徑均約23~25 nm,且離心穩(wěn)定性好。
[Abstract]:Based on the results of pre-test, the prescription composition of alprostadil microemulsion was first determined, and then the optimum dosage of each component in the formulation was determined by the method of star design-effect surface and drawing the curve of conductivity and water content.Finally, the stirring speed, time and temperature of the microemulsion were optimized according to the particle size and drug content.The results showed that the optimized formulation of alprostadil microemulsion was: 1 Solutol HS 158, egg yolk lecithin 2, medium chain triglyceride 4, glycerol 6% and water 80%, and the stirring speed, temperature and time were 300 r / min 30 鈩，
