米諾環(huán)素對大鼠腦缺血再灌注損傷后學習記憶能力及海馬神經(jīng)元PSD-95蛋白表達的影響

【摘要】:目的:探討靜脈用低劑量米諾環(huán)素對局灶性腦缺血再灌注大鼠認知功能障礙及缺血側(cè)海馬生長相關(guān)蛋白-43(growth associated protein-43,GAP-43)和突觸后致密物質(zhì)-95(postsynaptic density 95,PSD-95)表達的影響。方法:采用線栓法建立Sprague-Dawley大鼠大腦中動脈阻塞再灌注模型,將45只雄性大鼠隨機分為假手術(shù)組、模型組及米諾環(huán)素組(n=15)。分別采用免疫組織化學及免疫印跡法檢測術(shù)后2周大鼠缺血側(cè)海馬GAP-43和PSD-95蛋白的表達,Morris水迷宮實驗評價大鼠的行為學改變。結(jié)果:同假手術(shù)組相比,模型組大鼠缺血側(cè)海馬GAP-43(0.49±0.03)和PSD-95(0.92±0.04)表達明顯增高(P=0.000);大鼠逃避潛伏期明顯延長[(44.2±10.0)s],穿過原平臺次數(shù)(2.6±0.9)和在目標象限探索時間百分率明顯降低[(19.2±2.1)%,P=0.000)]。同模型組相比,尾靜脈給予3 mg/kg米諾環(huán)素治療后,大鼠缺血側(cè)海馬GAP-43(0.72±0.05)表達明顯增加,PSD-95表達降低(0.67±0.05,P=0.000),大鼠逃避潛伏期明顯縮短[(30.8±7.6)s,P=0.020]、穿過原平臺次數(shù)(4.4±1.1,P=0.012)和在目標象限探索時間百分率明顯增加[(30.4±2.7)%,P=0.000)]。結(jié)論:低劑量米諾環(huán)素能顯著改善腦缺血再灌注后大鼠學習記憶能力,其機制可能與上調(diào)GAP-43和下調(diào)PSD-95表達有關(guān)。
[Abstract]:Aim: to investigate the effects of low dose minocycline on cognitive impairment and the expression of hippocampal growth-associated protein -43 associated protein-43 (GAP-43) and postsynaptic postsynaptic density 95 (PSD-95) in rats with focal cerebral ischemia-reperfusion injury.Methods: the middle cerebral artery occlusion reperfusion model of Sprague-Dawley rats was established by thread embolization. 45 male rats were randomly divided into sham-operation group, model group and minocycline group.The expressions of GAP-43 and PSD-95 protein in hippocampus of ischemic rats were detected by immunohistochemistry and Western blotting at 2 weeks after operation.
