氣相色譜-質(zhì)譜法測定大鼠血漿中尼古丁及其藥物代謝動力學(xué)研究

【摘要】:采用氣相色譜-質(zhì)譜法測定大鼠血漿中的尼古丁。血漿樣品采用蛋白沉淀法凈化,采用DB-Wax毛細(xì)管色譜柱分離,質(zhì)譜中選擇電子轟擊離子源和選擇離子監(jiān)測模式,以茴香腦為內(nèi)標(biāo)進行定量。并采用PKSlover 2.0軟件計算藥代動力學(xué)參數(shù)。尼古丁的線性范圍為0.10~2.0mg·L-1,檢出限為4.59μg·L-1。對空白樣品進行加標(biāo)回收試驗,回收率在85.0%~90.1%之間,相對標(biāo)準(zhǔn)偏差(n=6)小于3.0%。應(yīng)用此方法考察了灌胃給藥的大鼠血漿樣品中尼古丁代謝動力學(xué)特點,主要動力學(xué)參數(shù)AUC0-t,AUC0-∞,Tmax和ρmax分別為8.570mg·L-1·h-1,10.49mg·L-1·h-1,1.0h,998.9μg·L-1,結(jié)果呈現(xiàn)藥物代謝動力學(xué)過程。
[Abstract]:Nicotine in rat plasma was determined by gas chromatography-mass spectrometry. Plasma samples were purified by protein precipitation and separated by DB-Wax capillary column. Electron bombardment ion source and ion monitoring mode were selected in mass spectrometry. PKSlover 2.0 software was used to calculate the pharmacokinetic parameters. The linear range of nicotine was 0.10~2.0mg L -1 and the detection limit was 4.59 渭 g L -1. The recovery rate of the blank sample was 85.0%, and the recovery rate was 90.1%.
