螺旋藻激酶對動脈粥樣硬化模型大鼠血管內(nèi)皮功能的影響

【摘要】:目的:研究螺旋藻激酶(SPK)對動脈粥樣硬化模型大鼠血管內(nèi)皮功能的影響。方法:將60只大鼠隨機分為正常對照組(蒸餾水)、模型組(蒸餾水)、陽性對照組(辛伐他汀,0.005 g/kg)和SPK低、中、高劑量組(80、160、320 U/kg)。除正常對照組外,其余各組大鼠均復(fù)制動脈粥樣硬化模型;同時,各組大鼠ig相應(yīng)藥物,每天1次,連續(xù)給藥12周。測定大鼠血清中總膽固醇(TC)、三酰甘油(TG)、低密度脂蛋白膽固醇(LDL-C)、高密度脂蛋白膽固醇(HDL-C)、白細胞介素6(IL-6)和腫瘤壞死因子α(TNF-α)的含量;蘇木精-伊紅染色觀察大鼠胸主動脈血管內(nèi)膜的形態(tài)改變。結(jié)果:與正常對照組比較,模型組大鼠血清中TC、TG、LDL-C、IL-6和TNF-α含量增加(P0.01),HDL-C含量降低(P0.01);血管內(nèi)皮細胞脫落,內(nèi)膜增生、向管腔內(nèi)突起,平滑肌細胞增殖且排列紊亂,中膜彈力纖維崩解斷裂。與模型組比較,各給藥組大鼠血清中TC、TG、LDL-C、IL-6和TNF-α含量下降(P0.05或P0.01),陽性對照組和SPK中、高劑量組大鼠血清中HDL-C含量增加(P0.05);給藥組大鼠血管內(nèi)皮細胞形態(tài)較模型組明顯改善,其中SPK中、高劑量組大鼠血管內(nèi)皮細胞各層結(jié)構(gòu)完整、內(nèi)膜基本光滑,SPK中劑量組大鼠血管中膜平滑肌細胞排列稍紊亂,與正常對照組比較無明顯變化。結(jié)論:SPK有明顯的降脂和抗炎作用,可保護血管內(nèi)皮功能;其作用機制可能與降低血清中TC、TG、LDL-C、IL-6、TNF-α含量和升高血清中HDL-C含量有關(guān)。
[Abstract]:Objective: to study the effect of Spirulina kinase (SPK) on vascular endothelial function in atherosclerotic rats. Methods: 60 rats were randomly divided into normal control group (distilled water group), model group (distilled water group) and positive control group (Simvastatin group). Statins 0.005 g / kg) and SPK low, In the middle and high dose groups, the atherosclerosis model was established in all the rats except the normal control group, and the rats in each group were given corresponding drugs once a day. For 12 weeks, the contents of total cholesterol (TCN), triglyceride (TGN), low density lipoprotein cholesterol (LDL-CU), high density lipoprotein cholesterol (HDL-CU), interleukin-6 (IL-6) and tumor necrosis factor- 偽 (TNF- 偽) in serum of rats were determined. Results: compared with the normal control group, the serum levels of TCU, TGG, LDL-CU, IL-6 and TNF- 偽 in the model group increased the content of HDL-C, decreased the content of P0.01C and decreased the content of P0.01C, the vascular endothelial cells fell off and the intima proliferated. The smooth muscle cells proliferated and arranged in disorder, and the elastic fibers were disintegrated and broken. Compared with the model group, the contents of IL-6 and TNF- 偽 in the serum of the rats in each group were decreased by P0.05 or P0.01a, and those in the positive control group and SPK were lower than those in the control group. The content of HDL-C in serum of rats in high dose group increased P0.05, and the morphology of vascular endothelial cells in the treatment group was significantly improved than that in the model group. In the SPK group, the structure of each layer of vascular endothelial cells in the high dose group was intact. Compared with the normal control group, the vascular smooth muscle cells in the middle dose group were slightly disordered, but there was no obvious change. Conclusion: SPK has obvious antilipidemic and anti-inflammatory effects, and can protect the vascular endothelial function. The mechanism may be related to the decrease of serum TCG-TG-LDL-Con IL-6 TNF- 偽 content and the increase of serum HDL-C content.
