【摘要】：吡唑作為重要的有機(jī)雜環(huán)化合物因其結(jié)構(gòu)的多樣性和良好的生物活性近年來逐漸引起廣大化學(xué)家的關(guān)注。大量研究和實(shí)踐表明吡唑類化合物具有抗菌、抗痙攣、消炎、植物生長(zhǎng)調(diào)節(jié)和抗血小板凝聚等生物活性。吡唑類化合物在農(nóng)藥領(lǐng)域作為植物保護(hù)性殺菌劑、除草劑、殺蟲劑、除螨劑,在醫(yī)藥領(lǐng)域作為抗菌素、抗血栓藥等方面有廣泛的應(yīng)用。 我們發(fā)展了一種DABCO促進(jìn)的由對(duì)甲苯磺酰腙和硝基烯制備多取代吡唑的方法。較之前報(bào)道的重氮化合物與缺電子烯烴或炔烴的1,3-偶極環(huán)加成反應(yīng),該反應(yīng)沒有經(jīng)過重氮中間體,而以連續(xù)的Baylis-Hillman/分子內(nèi)環(huán)化機(jī)制進(jìn)行,并且反應(yīng)的區(qū)域選擇性得到了翻轉(zhuǎn),為多取代吡唑的合成提供了一條新穎、有效的途徑。 本論文主要分為三章。第一章綜述了吡唑及其衍生物的應(yīng)用,以及各種制備方法。第二章詳細(xì)介紹了對(duì)甲苯磺酰腙與硝基烯在DABCO作用下合成吡唑類化合物的過程,同時(shí)對(duì)其反應(yīng)機(jī)理進(jìn)行了討論。第三章實(shí)驗(yàn)部分是具體的實(shí)驗(yàn)方法以及波普數(shù)據(jù)。
[Abstract]:As an important organic heterocyclic compound, pyrazole has attracted the attention of chemists in recent years because of its diversity of structure and good biological activity. A large number of studies and practices have shown that pyrazole compounds have antibacterial, anti-spasmodic and anti-inflammatory properties. Pyrazole compounds are used as plant protective fungicides, herbicides, insecticides, mites, and antimicrobial agents in the field of medicine. Antithrombotic agents are widely used in many fields. We have developed a DABCO promoted method for the preparation of polysubstituted pyrazole from p-toluenesulfonyl hydrazone and nitrene. The diazo compounds have been reacted with 1 / 3 dipolar cycloaddition of electron-deficient olefins or alkynes without diazo intermediates. The intramolecular cyclization of Baylis-Hillman/ was carried out continuously, and the region selectivity of the reaction was reversed, which provided a novel and effective way for the synthesis of polysubstituted pyrazole. In chapter 1, the application of pyrazole and its derivatives and various preparation methods are reviewed. In chapter 2, the synthesis of pyrazole compounds with p-toluenesulfonyl hydrazone and nitroene under the action of DABCO is introduced in detail. At the same time, the reaction mechanism is discussed. In chapter 3, the experimental method and Pope data are discussed.
