紫杉醇微膠囊的制備及其靶向釋放性能評價(jià)

【摘要】:以銀杏辛烯基琥珀酸淀粉酯為壁材,紫杉醇為芯材,采用噴霧干燥法制備微膠囊。通過正交試驗(yàn)確定了噴霧干燥法制備紫杉醇微膠囊的最優(yōu)工藝組合:紫杉醇與銀杏辛烯基琥珀酸淀粉酯質(zhì)量比例1∶8,乳狀液固形物質(zhì)量分?jǐn)?shù)15.0%,均質(zhì)壓力40.0 MPa,噴霧干燥進(jìn)風(fēng)溫度200℃。在此工藝條件下,紫杉醇包埋率為96.82%,微膠囊中紫杉醇質(zhì)量分?jǐn)?shù)為10.63%。產(chǎn)品氣味純正,沒有明顯的苦味,顆粒表面平整光滑,細(xì)小均勻,具有良好的流散性,包埋效果好。在模擬人工胃液中,90.0 min,累積溶出率4.27%,基本不釋藥;在模擬人工小腸液中,紫杉醇4.0 h的累積溶出率25.6%;在模擬人工大腸環(huán)境中,12.0 h的體外累積溶出率達(dá)89.26%,24.0 h的體外累積溶出率達(dá)95.34%。表明紫杉醇微膠囊具有抗酸、抗消化、在大腸中易降解等特性,達(dá)到了大腸定位釋藥的效果,且其制備工藝簡單、可行,是一種有潛力用于提高紫杉醇的藥效并且降低不良反應(yīng)的緩釋新劑型。

The optimum technology for the preparation of paclitaxel microcapsules by spray drying method was determined by orthogonal test: the mass ratio of paclitaxel to Ginkgo biloba octenyl succinate starch ester was 1: 8, and the emulsion solid substance was obtained.
