阿奇霉素對(duì)大鼠胃排空及血漿胃動(dòng)素影響的研究

【摘要】:目的: 觀察大鼠采用不同劑量阿奇霉素灌胃后,全胃半排空時(shí)間及血漿胃動(dòng)素濃度的變化,揭示阿奇霉素致胃腸道癥狀的可能機(jī)制。 方法: 80只雄性wistar大鼠隨機(jī)分為對(duì)照組(8只)和觀察組(72只);觀察組按給藥劑量分為小劑量、中等劑量、大劑量組,每組按給藥時(shí)間分為1d組、3d組、6d組。觀察組大鼠采用阿奇霉素灌胃,小劑量組、中等劑量組、大劑量組各組灌藥劑量分別為3mg/100g、6mg/100g、12mg/100g。所有大鼠實(shí)驗(yàn)當(dāng)天先用SPECT行胃排空檢測(cè),試餐為含有99mTc-DTPA的牛奶,用專用軟件分析原始采集數(shù)據(jù),得到全胃半排時(shí)間(GET1/2)、50分鐘末殘留率百分比、胃排空曲線。胃排空檢測(cè)結(jié)束后,經(jīng)腹主動(dòng)脈采集動(dòng)脈血2m1,運(yùn)用放免法測(cè)定血漿胃動(dòng)素濃度。用統(tǒng)計(jì)軟件SPSS17.0進(jìn)行統(tǒng)計(jì)分析,計(jì)數(shù)資料以“均數(shù)±標(biāo)準(zhǔn)差”表示,p0.05為差異有顯著性,p0.01為差異有極顯著性意義。 結(jié)果: 小劑量組、中等劑量組、大劑量組全胃半排時(shí)間明顯縮短,與正常對(duì)照組(64.21±7.24)min比較,有統(tǒng)計(jì)學(xué)意義(p0.05)。組內(nèi)不同時(shí)間點(diǎn)比較無(wú)統(tǒng)計(jì)學(xué)意義(p0.05)。觀察組血漿胃動(dòng)素濃度,小劑量組與正常對(duì)照組比較無(wú)統(tǒng)計(jì)學(xué)意義(p0.05);中劑量組、大劑量組與正常對(duì)照組比較明顯增高,差異有統(tǒng)計(jì)學(xué)意義(p0.05)。 結(jié)論: 阿奇霉素可促進(jìn)胃排空。阿奇霉素促進(jìn)胃排空的機(jī)制可能為小劑量下僅為胃動(dòng)素樣作用;中等劑量、大劑量時(shí)除胃動(dòng)素樣作用外,有內(nèi)源性胃動(dòng)素參與。
[Abstract]:Objective:. The changes of total gastric emptying time and plasma motilin concentration after different doses of azithromycin were observed in rats to reveal the possible mechanism of gastrointestinal symptoms induced by azithromycin. Methods:. 80 male wistar rats were randomly divided into control group (n = 8) and observation group (n = 72). Each group was divided into 1 day group, 3 d group and 6 d group according to the time of administration. Rats in the observation group were given azithromycin intragastric perfusion, small dose group, middle dose group, high dose group and large dose group, the dosage of each group was 3 mg / 100 mg / 100 mg / 100 mg / 12 mg / 100 g respectively. All rats were tested for gastric emptying with SPECT on the day of the experiment. The test meal consisted of milk containing 99mTc-DTPA. By analyzing the original data with special software, the percentage of residual rate at the end of 50 minutes and the gastric emptying curve were obtained. After the detection of gastric emptying, the percentage of residual rate at the end of 50 minutes was obtained. The plasma motilin concentration was measured by radioimmunoassay (RIA). The count data were statistically analyzed by SPSS17.0. There was significant difference in the count data with "mean 鹵standard deviation" (P 0.05). Results:. Compared with the control group (64.21 鹵7.24 min), there was statistical significance (P 0.05). There was no significant difference between the two groups at different time points (P 0.05). The plasma motilin concentration in the observation group was significantly shorter than that in the control group (P < 0.05). There was no significant difference between the low dose group and the normal control group (P 0.05), while in the middle dose group, the large dose group was significantly higher than that in the normal control group, and the difference was statistically significant (P 0.05). Conclusion:. Azithromycin can promote gastric emptying. The mechanism of azithromycin promoting gastric emptying may be the action of motilin at low dose and endogenous motilin at middle dose and high dose.
